Enclot 500 mg Capsule is an antifungal medication containing Clotrimazole, primarily used for the treatment of vaginal yeast infections (vaginal candidiasis). It works by stopping the growth of fungi, providing relief from symptoms like itching, burning, and discharge.
Uses & Indications
Dosage
Adults
One 500 mg vaginal capsule, administered as a single dose, preferably at bedtime. Insert deeply into the vagina.
Elderly
No specific dosage adjustment required for elderly patients.
Renal_impairment
No specific dosage adjustment required for renal impairment, as systemic absorption is minimal.
How to Take
Insert the vaginal capsule deeply into the vagina, preferably at bedtime. An applicator may be provided for easier insertion. Do not take orally.
Mechanism of Action
Clotrimazole inhibits the biosynthesis of ergosterol, an essential component of the fungal cell membrane. This leads to structural and functional impairment of the cytoplasmic membrane, resulting in leakage of intracellular components and eventual fungal cell death.

Contraindications

Drug Interactions
Tacrolimus and Sirolimus
Clotrimazole, when administered systemically or topically with significant absorption, may interact with systemic tacrolimus or sirolimus, potentially increasing their plasma concentrations. This is less likely with minimal systemic absorption from vaginal use, but monitoring blood levels is advisable if used concomitantly.

Store in a cool, dry place, below 30°C. Protect from light and moisture. Keep out of reach of children.
Overdose
Overdose from vaginal administration is highly unlikely due to minimal systemic absorption. In case of accidental oral ingestion, gastric lavage and symptomatic supportive measures may be considered, although severe systemic effects are not expected due to low oral bioavailability.
Pregnancy & Lactation
Pregnancy: Category B. Generally considered safe for use during pregnancy, especially after the first trimester, under medical supervision. Lactation: Small amounts may be excreted in breast milk, but considered compatible with breastfeeding due to minimal systemic absorption and low potential for infant exposure.
