Fuclav 125 mg Tablet is a combination antibiotic containing Cefuroxime Axetil and Clavulanic Acid, used to treat a wide range of bacterial infections.
Uses & Indications
Dosage
Adults
Typically, one tablet (Cefuroxime 250 mg + Clavulanic Acid 125 mg) twice daily for 7-10 days, depending on the severity of infection.
Elderly
No specific dose adjustment is usually required unless there is severe renal impairment. Careful monitoring is advised.
Renal_impairment
Dosage adjustment is necessary for patients with severe renal impairment (creatinine clearance <30 mL/min). Consult a physician.
How to Take
Take orally, preferably with food, to enhance absorption and reduce gastrointestinal discomfort. Do not crush or chew the tablet.
Mechanism of Action
Cefuroxime, a second-generation cephalosporin, inhibits bacterial cell wall synthesis. Clavulanic acid inactivates beta-lactamase enzymes produced by bacteria, preventing cefuroxime degradation and extending its antibacterial spectrum.
Pharmacokinetics
Onset
Relatively rapid, usually within 1-2 hours.
Excretion
Both cefuroxime and clavulanic acid are primarily excreted by the kidneys, through glomerular filtration and tubular secretion.
Half life
Cefuroxime: approximately 1.2-1.5 hours. Clavulanic acid: approximately 1 hour.
Absorption
Cefuroxime axetil is absorbed from the gastrointestinal tract and rapidly hydrolyzed to cefuroxime. Clavulanic acid is also well absorbed orally. Food enhances the absorption of cefuroxime axetil.
Metabolism
Cefuroxime is not significantly metabolized in the body. Clavulanic acid is extensively metabolized and then eliminated.
Side Effects
Contraindications
- •Known hypersensitivity to cefuroxime, clavulanic acid, any other cephalosporins, penicillins, or any component of the formulation.
- •History of jaundice/hepatic dysfunction associated with cefuroxime + clavulanate therapy.
Drug Interactions
Probenecid
Increases plasma concentration and prolongs the half-life of cefuroxime.
Oral Anticoagulants
May potentiate the anticoagulant effect, requiring close monitoring of INR.
Oral Contraceptives
May reduce the efficacy of oral contraceptives; advise using alternative contraception.
Storage
Store below 30°C in a dry place, away from direct sunlight and moisture. Keep out of reach of children.
Overdose
In case of overdose, gastric lavage may be considered. Symptomatic and supportive treatment should be given. Hemodialysis can remove cefuroxime and clavulanic acid from the circulation.
Pregnancy & Lactation
Use during pregnancy only if clearly needed and potential benefits outweigh risks. Small amounts of cefuroxime are excreted in breast milk; use with caution during lactation. Consult a doctor.
