Description
Overview of the medicine
Omeprazole is a proton pump inhibitor used to treat acid-related conditions such as GERD, peptic ulcers, and Zollinger-Ellison syndrome.
Uses & Indications
Dosage
Adults
For GERD: 20 mg once daily for 4-8 weeks. For PUD: 20-40 mg once daily for 4-8 weeks. For Zollinger-Ellison Syndrome: Initial 60 mg once daily, adjust as needed.
Elderly
No specific dose adjustment needed, but caution with severe hepatic impairment.
Renal_impairment
No dose adjustment required.
How to Take
Take whole capsule with water, preferably in the morning before food. Do not chew or crush the capsule.
Mechanism of Action
Irreversibly blocks the H+/K+-ATPase (proton pump) in gastric parietal cells, thereby reducing gastric acid secretion.

Onset
Within 1 hour, maximal effect within 2 hours.
Excretion
Primarily via urine (approx 77%), remainder via feces.
Half life
Approximately 0.5-1 hour (plasma), but acid suppression effect lasts over 24 hours.
Absorption
Rapidly absorbed, bioavailability approx 30-40% (increases with repeat dosing).
Metabolism
Extensively metabolized in the liver by CYP2C19 and CYP3A4.

- Concomitant use with nelfinavir.
