Description
Overview of the medicine
Lutisone 0.05 mg Nasal Suspension is a corticosteroid nasal spray used to relieve symptoms of allergic rhinitis, such as sneezing, runny nose, nasal itching, and congestion. It works by reducing inflammation in the nasal passages.
Uses & Indications
Dosage
Adults
Adults and adolescents 12 years and older: Initially 2 sprays in each nostril once daily. Once symptoms are controlled, dose may be reduced to 1 spray in each nostril once daily. Maximum 2 sprays in each nostril daily.
Elderly
No specific dosage adjustment is required for elderly patients. Use adult dosage.
Renal_impairment
No dosage adjustment is necessary for patients with renal impairment.
How to Take
For intranasal use only. Shake the bottle gently before each use. Prime the pump before first use or if not used for several days. Blow the nose gently to clear nostrils. Close one nostril and insert the nozzle into the other nostril, pointing slightly away from the septum. While sniffing gently, press the pump once. Repeat for the other nostril.
Mechanism of Action
Lutisone, a synthetic corticosteroid, exerts its action by binding to glucocorticoid receptors. This leads to the inhibition of inflammatory mediators such as prostaglandins, leukotrienes, and cytokines. It reduces inflammation, swelling, and mucus production in the nasal passages, thereby alleviating allergic rhinitis symptoms.
Pharmacokinetics
Onset
Symptomatic improvement may begin within a few hours, but full therapeutic effect may take several days (typically 3-4 days).
Excretion
Excreted primarily via the feces (approximately 90%) and to a lesser extent via urine.
Half life
Systemic half-life is approximately 1.5 to 2 hours, but local effects persist longer.
Absorption
Minimal systemic absorption following intranasal administration, with bioavailability typically less than 2%.
Metabolism
Extensively metabolized in the liver, primarily via CYP3A4 enzymes, to inactive metabolites.
Side Effects
Contraindications
- Hypersensitivity to Lutisone or any components of the formulation.
- Untreated localized infection involving the nasal mucosa (e.g., herpes simplex).
- Recent nasal surgery or trauma, until healing has occurred.
Drug Interactions
CYP3A4 Inhibitors (e.g., Ritonavir, Ketoconazole)
Co-administration with potent CYP3A4 inhibitors may increase systemic exposure to Lutisone. Use with caution, and consider reducing the Lutisone dose or avoiding concomitant use if possible.
Storage
Store below 30°C in a dry place, away from direct sunlight. Do not freeze. Keep out of reach of children.
Overdose
Acute overdose with Lutisone is unlikely due to the low systemic absorption from the nasal route. Chronic overdose or exceeding recommended doses may lead to systemic corticosteroid effects, such as Cushing's syndrome or adrenal suppression. Treatment should be symptomatic and supportive.
Pregnancy & Lactation
Pregnancy Category C. Use in pregnancy only if the potential benefit justifies the potential risk to the fetus. It is not known whether Lutisone is excreted in human milk. Exercise caution when administered to a nursing mother. Consult a physician before use during pregnancy or lactation.
