Pregabalin is an anticonvulsant and neuropathic pain agent used to treat various conditions including neuropathic pain, fibromyalgia, generalized anxiety disorder, and as an adjunctive therapy for partial-onset seizures.
Uses & Indications
Dosage
Adults
Typical starting dose is 75 mg twice daily or 50 mg three times daily. May be increased to 150 mg twice daily or 100 mg three times daily within a week, based on response and tolerability. Maximum dose usually 600 mg/day.
Elderly
Dosage adjustment may be necessary in elderly patients with reduced renal function. Start with a lower dose.
Renal_impairment
Dosage must be adjusted based on creatinine clearance (CrCl). For CrCl 30-60 mL/min, maximum dose 300 mg/day; for CrCl 15-30 mL/min, maximum dose 150 mg/day; for CrCl <15 mL/min, maximum dose 75 mg/day. Hemodialysis patients require a supplementary dose after each dialysis session.
How to Take
Take orally with or without food. Swallow the capsule whole with water. Do not crush, chew, or open the capsule.
Mechanism of Action
Pregabalin binds with high affinity to the alpha2-delta (α2δ) subunit of voltage-gated calcium channels in the central nervous system, reducing the release of several neurotransmitters, including glutamate, noradrenaline, and substance P. This action is thought to stabilize hyperexcited neurons and reduce nerve pain signals.
