Description
Overview of the medicine
Pregabalin is an anticonvulsant and neuropathic pain agent used to treat neuropathic pain (e.g., diabetic neuropathy, postherpetic neuralgia), fibromyalgia, and as adjunctive therapy for partial onset seizures in adults.
Uses & Indications
Dosage
Adults
Initial dose 75 mg twice daily or 50 mg three times daily (150 mg/day). May be increased gradually up to 150-600 mg/day in divided doses, depending on indication and patient response.
Elderly
Lower doses may be required due to age-related decrease in renal function. Start with 25-50 mg daily and titrate slowly.
Renal_impairment
Dose adjustment necessary based on creatinine clearance. For creatinine clearance <30 mL/min, dose should be reduced significantly.
How to Take
Lyric capsules should be taken orally with or without food. Swallow the capsule whole with water.
Mechanism of Action
Pregabalin binds with high affinity to the alpha-2-delta (α2-δ) subunit of voltage-gated calcium channels in the central nervous system, which is thought to modulate the release of several excitatory neurotransmitters.

Onset
Variable; pain relief can start within a week, but full therapeutic effect may take several weeks.
Excretion
Primarily excreted unchanged in the urine; renal elimination is the major route of excretion.
Half life
Approximately 6.3 hours.
Absorption
Rapidly absorbed after oral administration, peak plasma concentrations usually occur within 1 hour. Oral bioavailability is >90% and is not affected by food.
Metabolism
Undergoes negligible metabolism in humans. Less than 0.1% of the dose is excreted in the urine as metabolites.
