Maprocin 500 mg Tablet is a broad-spectrum macrolide antibiotic used to treat a variety of bacterial infections.
Uses & Indications
Dosage
Adults
500 mg once daily for 3 days, or 500 mg on day 1 followed by 250 mg once daily on days 2-5.
Elderly
No dosage adjustment required.
Renal_impairment
No dosage adjustment required for mild to moderate renal impairment. Use with caution in severe impairment.
How to Take
Maprocin tablets can be taken orally with or without food. For best absorption, it is often recommended to take it an hour before or two hours after meals, though this is less critical than for some other macrolides.
Mechanism of Action
Azimaprocin acts by inhibiting bacterial protein synthesis. It binds to the 50S ribosomal subunit of susceptible microorganisms, thereby interfering with the translocation of peptides and inhibiting RNA-dependent protein synthesis.
Pharmacokinetics
Onset
Peak plasma concentrations reached within 2-3 hours.
Excretion
Mainly excreted unchanged in bile, with a small amount excreted via urine.
Half life
Approximately 48-72 hours, due to extensive tissue uptake and slow release.
Absorption
Rapidly absorbed from the gastrointestinal tract; bioavailability is about 37%.
Metabolism
Primarily hepatic, via demethylation.
Side Effects
Contraindications
- Hypersensitivity to azimaprocin, erythromycin, or any macrolide or ketolide antibiotic.
- History of cholestatic jaundice/hepatic dysfunction associated with prior azimaprocin use.
Drug Interactions
Warfarin
Increased anticoagulant effect (INR) has been reported. Monitor INR closely.
Nelfinavir
Increased serum concentrations of azimaprocin. Monitor for side effects.
Antacids (containing aluminum or magnesium)
May reduce the absorption of azimaprocin. Take azimaprocin at least 1-2 hours before or after antacids.
Storage
Store below 30°C in a dry place, away from direct light and moisture. Keep out of reach of children.
Overdose
In case of overdose, gastric lavage and general supportive measures are indicated. Hemodialysis and peritoneal dialysis are not effective in removing azimaprocin from the circulation.
Pregnancy & Lactation
Pregnancy Category B. Studies in animals have shown no evidence of harm to the fetus. However, there are no adequate and well-controlled studies in pregnant women. Use during pregnancy only if clearly needed. Azimaprocin is excreted into breast milk; therefore, caution should be exercised when administered to a nursing mother.
