Description
Overview of the medicine
Neurega-75 mg capsule contains Pregabalin, which is used to treat neuropathic pain, epilepsy (partial seizures), and generalized anxiety disorder. It works by reducing the release of certain neurotransmitters in the brain.
Uses & Indications
Dosage
Adults
Initial dose is typically 75 mg twice daily or 50 mg three times daily, which may be increased based on response and tolerability, up to a maximum of 600 mg/day.
Elderly
Dose adjustment may be necessary in elderly patients due to potential age-related decrease in renal function.
Renal_impairment
Dosage must be reduced in patients with impaired renal function based on creatinine clearance. For CrCl 30-60 mL/min, 75 mg twice daily or 50 mg three times daily (max 300 mg/day); for CrCl 15-30 mL/min, 75 mg once daily or 25-50 mg twice daily (max 150 mg/day).
How to Take
Take orally with or without food. Swallow the capsule whole with water.
Mechanism of Action
Pregabalin binds to the alpha-2-delta subunit of voltage-gated calcium channels in the central nervous system, which leads to a reduction in the release of various excitatory neurotransmitters such as glutamate, noradrenaline, and substance P. This action helps to reduce neuropathic pain and seizure activity.
Pharmacokinetics
Onset
Onset of action for pain relief typically occurs within 1 week; for anxiety, within 1 week; for epilepsy, it varies.
Excretion
Primarily excreted unchanged by the kidneys. Renal clearance is directly proportional to creatinine clearance.
Half life
Approximately 6.3 hours.
Absorption
Rapidly absorbed after oral administration, peak plasma concentrations reached within 1 hour. Bioavailability is ≥90% and is not affected by food.
Metabolism
Negligible metabolism in humans; less than 0.1% of the dose is excreted in the urine as metabolites.
Side Effects
Contraindications
- Hypersensitivity to pregabalin or any component of the formulation.
- Severe allergic reactions (e.g., angioedema) to pregabalin.
Drug Interactions
Opioid Analgesics
Increased risk of respiratory depression and sedation.
Thiazolidinediones (e.g., rosiglitazone)
Potential for increased peripheral edema and weight gain.
CNS Depressants (e.g., benzodiazepines, alcohol)
Additive effects on central nervous system depression, leading to increased drowsiness and sedation.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose include somnolence, confusion, agitation, and rarely, coma. Treatment is supportive and may involve gastric lavage or emesis if recent ingestion. Hemodialysis is effective in removing pregabalin.
Pregnancy & Lactation
Pregnancy Category C. Should be used during pregnancy only if the potential benefit justifies the potential risk to the fetus. Pregabalin is excreted in human milk; a decision should be made whether to discontinue nursing or to discontinue the drug, taking into account the importance of the drug to the mother.

YesClinical Trials
Extensive clinical trials have demonstrated the efficacy and safety of pregabalin in various indications including neuropathic pain, epilepsy, and generalized anxiety disorder. Ongoing research continues to explore its full therapeutic potential and safety profile.
Lab Monitoring
- Monitor renal function (creatinine clearance) periodically, especially in elderly or renally impaired patients, to adjust dosage.
- Monitor for signs of peripheral edema and weight gain.
Doctor Notes
- Adjust dosage carefully in patients with renal impairment based on creatinine clearance.
- Monitor patients for signs of suicidal ideation and behavior, especially at the initiation of therapy or after dose changes.
- Advise patients about the potential for central nervous system (CNS) depression, including somnolence and dizziness, and to avoid concomitant use with alcohol or other CNS depressants.
Patient Guidelines
- Do not stop taking Neurega-75 mg capsule abruptly without consulting your doctor, as this may lead to withdrawal symptoms.
- This medicine may cause dizziness or drowsiness; avoid driving or operating heavy machinery until you know how it affects you.
- Report any unusual mood changes, suicidal thoughts, or signs of allergic reactions (swelling of face, lips, tongue, or throat) to your doctor immediately.
Missed Dose Advice
If you miss a dose, take it as soon as you remember. If it is almost time for your next dose, skip the missed dose and continue with your regular dosing schedule. Do not take a double dose to make up for a missed one.
Driving Precautions
Neurega-75 mg capsule can cause dizziness, drowsiness, and blurred vision. Patients should be advised not to drive, operate complex machinery, or engage in other hazardous activities until they are certain that pregabalin does not adversely affect their ability to perform such tasks.
Lifestyle Advice
- Avoid alcohol consumption while taking this medicine, as it can increase drowsiness and dizziness.
- Maintain a healthy lifestyle, including regular exercise and a balanced diet, to help manage underlying conditions.
