Nixon 1 gm injection contains Ceftriaxone, a broad-spectrum third-generation cephalosporin antibiotic used to treat various bacterial infections including severe ones like meningitis, sepsis, and complicated urinary tract infections.
Uses & Indications
Dosage
Adults
Typical dose: 1-2 gm once daily (IM or IV), up to 4 gm/day in severe infections.
Elderly
No dose adjustment typically required unless severe renal or hepatic impairment.
Renal_impairment
Generally no dose adjustment for mild to moderate impairment; for severe impairment, close monitoring is advised. Max 2gm/day in severe renal or hepatic impairment.
How to Take
Administer by deep intramuscular injection or by slow intravenous infusion (over 30 minutes) or slow intravenous injection (over 2-4 minutes). Reconstitute with appropriate diluents (e.g., sterile water for injection, lidocaine solution for IM).
Mechanism of Action
Ceftriaxone inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins (PBPs), leading to bacterial cell lysis and death.

Onset
Peak plasma concentration within 2-3 hours after IM injection; immediate after IV.
Excretion
Approximately 50-60% excreted unchanged in urine, and 40-50% in bile/feces.

Approximately 6-9 hours.
Absorption
Rapidly and completely absorbed after intramuscular (IM) injection. Bioavailability is 100% for IV and IM administration.
Metabolism
Limited hepatic metabolism; primarily forms inactive metabolites.
Side Effects
Contraindications
- •Hypersensitivity to ceftriaxone or any other cephalosporins
- •Neonates with hyperbilirubinemia, especially premature infants
- •Neonates requiring calcium-containing IV solutions (due to precipitation risk)
Drug Interactions
Aminoglycosides
Synergistic antibacterial activity, but potential for increased nephrotoxicity (rare).
Calcium-containing solutions
Concomitant administration is contraindicated in neonates and infants due to risk of precipitation.
Loop diuretics (e.g., Furosemide)
May increase nephrotoxicity, especially with other nephrotoxic drugs.
Oral anticoagulants (e.g., Warfarin)
Increased anticoagulant effect, requiring close monitoring of INR.
Storage
Store below 30°C in a dry place, away from light. Do not freeze. Reconstituted solution should be used immediately or within 24 hours if refrigerated.
Overdose
Symptoms of overdose may include nausea, vomiting, diarrhea, and central nervous system effects (e.g., convulsions) in susceptible individuals. Management is symptomatic and supportive; hemodialysis or peritoneal dialysis is not effective in removing ceftriaxone.
Pregnancy & Lactation
Pregnancy Category B. Studies in animals have shown no evidence of fetal harm, but adequate and well-controlled studies in pregnant women are lacking. Use during pregnancy only if clearly needed. Excreted in low concentrations in breast milk; caution is advised when administering to a nursing mother.
