Description
Overview of the medicine
Odafen (Ketotifen) is an antihistamine and mast cell stabilizer primarily used for the prophylactic treatment of bronchial asthma, allergic conjunctivitis, and other allergic conditions. It reduces allergic inflammatory responses.
Uses & Indications
Dosage
Adults
Adults: 1 mg twice daily with food. If necessary, the dose may be increased to 2 mg twice daily.
Elderly
Elderly: No special dosage recommendations are necessary.
Renal_impairment
Renal Impairment: No dosage adjustment is generally required.
How to Take
Administer orally with food to reduce GI upset. For full prophylactic effect on asthma, treatment should be continued for several weeks.
Mechanism of Action
Ketotifen acts as a selective H1-histamine receptor antagonist and a mast cell stabilizer. It inhibits the release of inflammatory mediators (like histamine, leukotrienes) from mast cells and basophils, thereby reducing allergic reactions.

Onset
Full therapeutic effect for asthma prophylaxis may take several weeks (2-12 weeks).
Excretion
Approximately 60-70% excreted in urine, 30-40% in feces.
Half life
Biphasic elimination with a terminal half-life of 21 hours.
Absorption
Well absorbed orally; bioavailability approx. 50% due to first-pass metabolism.
Metabolism
Extensively metabolized in the liver via N-demethylation and glucuronidation.
Side Effects
Contraindications
- •Hypersensitivity to Ketotifen or any excipients.
- •Not for use in acute asthma attacks; it is a prophylactic agent.
Drug Interactions
Oral Antidiabetics
Reversible fall in platelet count has been reported; avoid concurrent use or monitor platelet levels.
CNS Depressants and Alcohol
Ketotifen may potentiate the effects of sedatives, hypnotics, antihistamines, and alcohol.
Storage
Store below 30°C in a dry place, away from light and moisture.
Overdose
Symptoms of overdose include drowsiness, confusion, disorientation, tachycardia, and possibly coma. Treatment is symptomatic and supportive, including gastric lavage and activated charcoal.
Pregnancy & Lactation
Pregnancy Category C: Should only be used if the potential benefit justifies the potential risk to the fetus. Lactation: Ketotifen is excreted in human milk; avoid use during breastfeeding or discontinue breastfeeding.
