Onepro 20 mg Capsule contains Esomeprazole, a proton pump inhibitor used to reduce stomach acid production and treat conditions like GERD and ulcers.
Uses & Indications
Dosage
Adults
For GERD: 20 mg or 40 mg once daily for 4-8 weeks. For Erosive Esophagitis: 20 mg or 40 mg once daily for 4-8 weeks. For H. pylori Eradication: 20 mg twice daily for 10 days (in combination with antibiotics).
Elderly
No specific dose adjustment is required, but caution is advised due to potential age-related decrease in liver function.
Renal_impairment
No dosage adjustment is necessary for patients with impaired renal function.
How to Take
Take the capsule whole with water at least one hour before a meal. Do not chew or crush the capsule. If swallowing is difficult, the capsule can be opened and the contents mixed with a small amount of water or fruit juice (do not store the mixture).
Mechanism of Action
Esomeprazole selectively binds to and irreversibly inhibits the H+/K+-ATPase (proton pump) in the gastric parietal cells, thereby suppressing gastric acid secretion.

Excretion
Primarily via urine (approximately 80%) as metabolites and feces (approximately 20%).

Side Effects
Contraindications
- •Hypersensitivity to Esomeprazole, substituted benzimidazoles, or any component of the formulation.
- •Concomitant use with nelfinavir.
Drug Interactions
Warfarin
Increased INR and prothrombin time; monitor closely for bleeding.
Clopidogrel
May reduce the antiplatelet effect of clopidogrel (clinical significance debated); use with caution.
Methotrexate
Increased methotrexate levels, especially with high doses; consider temporary PPI discontinuation.
Nelfinavir, Atazanavir
Decreased plasma levels of antivirals due to increased gastric pH, leading to potential loss of virologic response.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms are generally mild and self-limiting (e.g., drowsiness, tachycardia, nausea, diarrhea, headache). Treatment is symptomatic and supportive; no specific antidote is available.
