Overview of the medicine
Onepro 40 mg Tablet is a proton pump inhibitor (PPI) used to reduce stomach acid production. It treats conditions like heartburn, gastroesophageal reflux disease (GERD), stomach ulcers, and Zollinger-Ellison syndrome.
Uses & Indications
Dosage
Adults
Typically 20-40 mg once daily, depending on the condition. For GERD, 20 mg once daily for 4-8 weeks. For duodenal ulcers, 20 mg once daily for 2-4 weeks. For gastric ulcers, 20 mg once daily for 4-8 weeks. For Zollinger-Ellison Syndrome, initial dose 60 mg once daily, adjusted as needed.
Elderly
No specific dose adjustment is generally required, but caution should be exercised.
Renal_impairment
No dose adjustment is needed for patients with impaired renal function.
How to Take
Take the tablet whole with a glass of water, preferably in the morning before food. Do not chew or crush.
Mechanism of Action
Omeprazole inhibits the H+/K+-ATPase enzyme system (the proton pump) in the gastric parietal cells, thereby blocking the final step of acid production.
Pharmacokinetics
Onset
Acid secretion inhibition occurs within 1 hour, maximal effect within 2 hours.
Excretion
Mainly excreted in urine (approx. 77%) and feces (approx. 18%).
Half life
Plasma half-life is approximately 0.5-1 hour, but duration of acid suppression is much longer (up to 72 hours) due to irreversible binding.
Absorption
Rapidly absorbed after oral administration, peak plasma concentrations reached within 0.5-3.5 hours.
Metabolism
Extensively metabolized in the liver, primarily by CYP2C19 and CYP3A4.

- Hypersensitivity to omeprazole or any components of the formulation
- Concomitant use with nelfinavir

Decreased metabolism and increased plasma levels of diazepam.
Warfarin
Increased INR/prothrombin time.
