Rovantin 100 mg Tablet contains Aceclofenac, a non-steroidal anti-inflammatory drug (NSAID), used for pain relief and reduction of inflammation in various conditions like osteoarthritis, rheumatoid arthritis, and ankylosing spondylitis.
Uses & Indications
Dosage
Adults
100 mg twice daily (morning and evening).
Elderly
No dosage adjustment is generally required, but caution should be exercised due to increased risk of adverse effects.
Renal_impairment
No dosage reduction needed for mild renal impairment; use with caution in moderate to severe impairment.
How to Take
Rovantin 100 mg tablet should be taken orally, preferably with or after food, with an adequate amount of liquid. Do not chew or crush the tablet.
Mechanism of Action
Aceclofenac exerts its anti-inflammatory, analgesic, and antipyretic effects by inhibiting prostaglandin synthesis, primarily through the inhibition of cyclooxygenase (COX) enzymes, particularly COX-2.
Pharmacokinetics
Onset
Within 30-60 minutes.
Excretion
Approximately two-thirds of the dose is excreted in the urine and one-third in the faeces, mainly as hydroxylated metabolites.
Half life
Approximately 4-4.3 hours.
Absorption
Rapidly and completely absorbed after oral administration, peak plasma concentrations reached in 1.25-3 hours.
Metabolism
Extensively metabolized in the liver, primarily by CYP2C9.
Side Effects
Contraindications
- •Hypersensitivity to aceclofenac or any excipients
- •Patients with active or history of recurrent peptic ulcer/haemorrhage
- •History of gastrointestinal bleeding or perforation related to previous NSAID therapy
- •Severe heart failure, hepatic failure, or renal failure
- •Third trimester of pregnancy
- •Patients in whom aspirin or other NSAIDs precipitate attacks of asthma, urticaria, or acute rhinitis
Drug Interactions
Diuretics
May reduce the natriuretic effect of diuretics.
Methotrexate
Increases plasma levels and toxicity of methotrexate.
Corticosteroids
Increased risk of gastrointestinal ulceration or bleeding.
Antihypertensives
May reduce the antihypertensive effect.
Lithium and Digoxin
Increases plasma concentration of lithium and digoxin.
Anticoagulants (e.g., Warfarin)
Increased risk of bleeding.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose may include nausea, vomiting, epigastric pain, gastrointestinal bleeding, diarrhoea, dizziness, disorientation, excitation, coma, drowsiness, tinnitus, and convulsions. Management is symptomatic and supportive, including gastric lavage or activated charcoal.
Pregnancy & Lactation
Not recommended during the third trimester of pregnancy due to potential for premature closure of the ductus arteriosus and inhibition of uterine contractions. Use during other trimesters or lactation only if potential benefit outweighs risk. Consult a doctor.
