Description
Overview of the medicine
Rovantin 200 mg Tablet contains Aceclofenac, a non-steroidal anti-inflammatory drug (NSAID), used for the relief of pain and inflammation in conditions like osteoarthritis, rheumatoid arthritis, and ankylosing spondylitis.
Uses & Indications
Dosage
Adults
The recommended dose is 100 mg twice daily. Rovantin 200 mg should be taken as directed by a physician, typically once daily or as advised.
Elderly
The lowest effective dose should be used, and patients should be monitored for potential adverse effects.
Renal_impairment
No dosage adjustment is generally required in mild renal impairment; however, caution is advised in moderate to severe impairment. Avoid in severe renal failure.
How to Take
Tablets should be swallowed whole with a sufficient amount of liquid, with or after food. Do not chew or crush.
Mechanism of Action
Aceclofenac primarily acts by inhibiting cyclooxygenase (COX) enzymes, particularly COX-2, which are involved in the synthesis of prostaglandins. Prostaglandins are mediators of pain, inflammation, and fever.
Pharmacokinetics
Onset
Pain relief typically begins within 30-60 minutes.
Excretion
Excreted mainly via urine (about 2/3) and feces (about 1/3) as metabolites.
Half life
Approximately 4 hours.
Absorption
Rapidly and completely absorbed from the gastrointestinal tract after oral administration, with peak plasma concentrations reached in about 1.25 to 3 hours.
Metabolism
Extensively metabolized in the liver, primarily by cytochrome P450 2C9 (CYP2C9) to 4'-hydroxyaceclofenac and other metabolites.
Side Effects
Contraindications
- •Hypersensitivity to aceclofenac or any component of the tablet
- •History of asthma, urticaria, or allergic-type reactions after taking aspirin or other NSAIDs
- •Active or history of recurrent peptic ulcer/hemorrhage
- •Severe heart failure, severe hepatic failure, or severe renal failure
- •Third trimester of pregnancy
Drug Interactions
SSRIs
Increased risk of gastrointestinal bleeding.
Corticosteroids
Increased risk of gastrointestinal ulceration or bleeding.
Diuretics and ACE inhibitors
Reduced hypotensive effect and increased risk of renal impairment.
Lithium, Digoxin, Methotrexate
Increased plasma levels, leading to toxicity.
Warfarin and other anticoagulants
Increased risk of bleeding.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms may include headache, nausea, vomiting, epigastric pain, gastrointestinal bleeding, rarely diarrhea, disorientation, excitation, coma, drowsiness, dizziness, tinnitus, fainting, and convulsions. Management involves symptomatic and supportive treatment, including gastric lavage or activated charcoal if ingested recently.
Pregnancy & Lactation
Avoid during the third trimester of pregnancy due to potential fetal harm (e.g., premature closure of ductus arteriosus). Use with caution in the first two trimesters and during lactation only if the potential benefit outweighs the risk, under medical supervision. It's generally recommended to avoid NSAIDs during breastfeeding.
