Uses & Indications
Dosage

Not typical dosage form for adults, usually higher strengths or different forms. If used, typically 250-500 mg every 8 hours.
Elderly
Dosage adjustment based on renal function.
Renal_impairment
Reduced dose and/or increased dosing interval based on creatinine clearance.
How to Take
Administer orally. Shake the suspension well before each use. Can be taken with or without food. Complete the full course of treatment as prescribed, even if symptoms improve.
Mechanism of Action
Amoxicillin is a beta-lactam antibiotic that inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins (PBPs), leading to bacterial cell lysis and death.
Pharmacokinetics
Onset
Within 30-60 minutes
Excretion
Primarily excreted unchanged in the urine via renal tubular secretion and glomerular filtration.
Half life
Approximately 1-1.5 hours (in adults with normal renal function)
Absorption
Rapidly and well absorbed from the gastrointestinal tract, stable in gastric acid. Bioavailability is approximately 70-90%.
Metabolism
Limited metabolism; about 10-25% is metabolized to inactive penicilloic acid.
Side Effects
Contraindications
- Known hypersensitivity to penicillin or any component of Amoxicillin
- Previous history of severe allergic reaction to other beta-lactam antibiotics (e.g., cephalosporins)
Drug Interactions
Warfarin
May increase the anticoagulant effect (monitor INR).
Probenecid
Increases and prolongs Amoxicillin blood levels.
Allopurinol
Increased risk of rash.
Methotrexate
May increase methotrexate toxicity.
Oral Contraceptives
May reduce the effectiveness of oral contraceptives.
Storage
Store dry powder below 30°C (86°F), protected from moisture. Store reconstituted suspension in a refrigerator (2-8°C/36-46°F). Do not freeze.
Overdose
Symptoms may include nausea, vomiting, diarrhea, abdominal pain, and rarely convulsions. Treatment is generally supportive and symptomatic. Gastric lavage or activated charcoal may be considered if recent ingestion. Hemodialysis can remove Amoxicillin from circulation.
Pregnancy & Lactation
Pregnancy Category B. Generally considered safe during pregnancy. Excreted in breast milk in small amounts; generally considered compatible with breastfeeding, but monitor infant for potential side effects like diarrhea or rash.
