Description
Overview of the medicine
Trialon 55 mcg Nasal Spray is a corticosteroid used to treat symptoms of allergic rhinitis, including sneezing, runny nose, nasal congestion, and itchy or watery eyes, in adults and children.
Uses & Indications
Dosage
Adults
Initially 2 sprays in each nostril once daily. Once symptoms are controlled, reduce to 1 spray in each nostril once daily.
Elderly
No dosage adjustment required.
Renal_impairment
No dosage adjustment required.
How to Take
Administer as a spray into each nostril. Shake gently before use. Clear nasal passages before spraying. Prime the pump by spraying into the air 6 times if not used for 30 days or if the cap has been left off.
Mechanism of Action
Fluticasone Furoate is a synthetic trifluorinated corticosteroid with potent anti-inflammatory activity. It works by reducing inflammation in the nasal passages, thereby alleviating symptoms of allergic rhinitis.
Pharmacokinetics
Onset
Initial effects seen within 8 hours, full effect within several days of consistent use.
Excretion
Primarily excreted in the feces, with less than 1% excreted in urine.
Half life
Approximately 15-20 hours (systemic elimination half-life).
Absorption
Low systemic absorption (typically less than 0.5%) due to extensive first-pass metabolism.
Metabolism
Extensive first-pass metabolism in the liver by the cytochrome P450 3A4 (CYP3A4) isoenzyme.
Side Effects
Contraindications
- Hypersensitivity to fluticasone furoate or any other components of the spray.
Drug Interactions
Ritonavir
Concurrent use with ritonavir (a strong CYP3A4 inhibitor) is not recommended due to significantly increased systemic exposure to fluticasone furoate, leading to potential systemic corticosteroid effects.
Other strong CYP3A4 inhibitors (e.g., Ketoconazole)
Use with caution; may increase fluticasone furoate systemic exposure.
Storage
Store below 30°C. Do not freeze. Keep the bottle in an upright position. Keep out of the reach and sight of children.
Overdose
Acute overdose is unlikely due to the low systemic absorption of fluticasone furoate from nasal administration. Chronic overdose may lead to systemic corticosteroid effects. Treatment is symptomatic and supportive.
Pregnancy & Lactation
Use during pregnancy or lactation only if the potential benefit to the mother justifies the potential risk to the fetus or infant. Consult a healthcare professional before use.

Doctor Notes
- Educate patients on the correct technique for administering nasal spray to ensure optimal drug delivery and minimize local side effects.
- Emphasize that the full therapeutic effect may not be immediate and consistent daily use is crucial for best outcomes.
- Monitor for signs of local adverse effects (e.g., nasal irritation, bleeding) and systemic effects (e.g., growth in children) during long-term therapy.
Patient Guidelines
- Read the patient leaflet carefully before use and follow the instructions precisely.
- Do not exceed the recommended dose; overuse can lead to adverse effects.
- It may take several days of consistent use to achieve maximum relief.
- Consult your doctor if symptoms do not improve after a few days or worsen.
Missed Dose Advice
If a dose is missed, take it as soon as you remember. If it is almost time for the next dose, skip the missed dose and resume your regular dosing schedule. Do not double the dose to make up for a missed one.
