Description
Overview of the medicine
Flux 500 mg capsule is an antibiotic belonging to the penicillin group. It is used to treat bacterial infections, particularly those caused by penicillinase-producing staphylococci, such as skin and soft tissue infections, bone and joint infections, and respiratory tract infections.
Uses & Indications
Dosage
Adults
Oral: 250-500 mg every 6 hours, usually taken 30-60 minutes before meals. For severe infections, dosage may be increased up to 1 g every 6 hours.
Elderly
Generally, adult dosage can be used, but renal function should be assessed, and dosage adjusted if impairment is present.
Renal_impairment
For creatinine clearance <10 mL/min, reduce dose to 250 mg every 8-12 hours.
How to Take
Take the capsule whole with a glass of water, preferably on an empty stomach (30-60 minutes before food) to ensure optimal absorption. Do not chew or crush the capsule.
Mechanism of Action
Flucloxacillin is a beta-lactam antibiotic that inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins (PBPs). This leads to osmotic instability and bacterial cell lysis. It is resistant to degradation by beta-lactamase enzymes produced by certain bacteria, making it effective against penicillinase-producing staphylococci.

Within 1-2 hours (therapeutic effect usually observed within 24-48 hours of starting treatment).
Excretion
Mainly excreted unchanged in the urine, with some biliary excretion.

Warfarin
May increase anticoagulant effect, requiring closer INR monitoring.
Probenecid
Increases and prolongs plasma levels of flucloxacillin by reducing renal tubular secretion.
Methotrexate
May increase methotrexate toxicity due to reduced renal clearance.
Oral Contraceptives
May reduce efficacy of oral contraceptives; advise additional barrier contraception.

Overdose symptoms may include nausea, vomiting, diarrhea, and electrolyte imbalance. In severe cases, neurotoxicity (e.g., convulsions) may occur, especially in patients with impaired renal function. Treatment is supportive and symptomatic; hemodialysis can remove flucloxacillin from the blood.

Small amounts are excreted in breast milk; generally considered compatible with breastfeeding, but monitor the infant for gastrointestinal disturbances or candidiasis.
