Description
Overview of the medicine
Nyclobate 0.05% Scalp Solution contains Clobetasol Propionate, a very potent corticosteroid, used for the short-term treatment of severe inflammatory skin conditions of the scalp such as psoriasis and seborrheic dermatitis, which have not responded to less potent corticosteroids.
Uses & Indications
Dosage
Adults
Apply a thin layer to the affected areas of the scalp twice daily, in the morning and at night, until improvement occurs. Treatment should be limited to 2 consecutive weeks and amounts greater than 50 g per week should not be used.
Elderly
No specific dosage adjustment is generally required, but caution should be exercised due to potentially thinner skin and increased systemic absorption risk.
Renal_impairment
No specific dosage adjustment is required for topical application due to minimal systemic absorption.
How to Take
For external use only. Apply a few drops or a thin layer of the solution to the affected areas of the scalp and rub in gently. Do not cover the treated area with bandages or other occlusive dressings unless directed by a doctor. Wash hands immediately after use.
Mechanism of Action
Clobetasol Propionate is a corticosteroid that exhibits anti-inflammatory, antipruritic, and vasoconstrictive properties. It works by inducing phospholipase A2 inhibitory proteins, thereby controlling the biosynthesis of potent mediators of inflammation such as prostaglandins and leukotrienes.
Pharmacokinetics
Onset
Within hours of application.
Excretion
Excreted renally (via urine) as inactive metabolites, and to a lesser extent, via bile.
Half life
Variable, typically short due to rapid metabolism.
Absorption
Minimal systemic absorption occurs when applied topically, especially when skin barrier is intact. Absorption increases with extensive use, occlusion, or application to damaged skin.
Metabolism
Primarily metabolized in the liver, similar to other corticosteroids.
Side Effects
Contraindications
- Hypersensitivity to Clobetasol Propionate or any components of the formulation
- Untreated scalp infections (bacterial, fungal, viral, parasitic)
- Rosacea, acne vulgaris, perioral dermatitis
- Pruritus without inflammation
Drug Interactions
Ritonavir
May increase systemic exposure to Clobetasol Propionate, use with caution.
Itraconazole
May increase systemic exposure to Clobetasol Propionate, use with caution.
Storage
Store below 30°C. Protect from light and moisture. Keep out of reach of children.
Overdose
Acute overdose is unlikely due to the topical route. However, prolonged or excessive use may lead to systemic effects such as Cushing's syndrome, hyperglycemia, or adrenal suppression. Treatment is symptomatic and supportive.
Pregnancy & Lactation
Pregnancy Category C. Use in pregnancy only if the potential benefit justifies the potential risk to the fetus. Caution advised during lactation; it is not known whether topical Clobetasol is excreted in human milk. Avoid applying to the breast area.

Lab Monitoring
- Not routinely required for short-term topical use. For extensive or prolonged use, monitoring of hypothalamic-pituitary-adrenal (HPA) axis suppression (e.g., plasma cortisol levels) may be considered.
Doctor Notes
- Emphasize short-term use (max 2 weeks) for severe conditions.
- Caution against use on the face, intertriginous areas, or in children without specialist advice.
- Monitor for signs of systemic absorption or local adverse effects during treatment.
Patient Guidelines
- Use only as directed by your doctor and for the prescribed duration.
- Do not use on the face, groin, or axillae.
- Avoid contact with eyes; rinse immediately with water if contact occurs.
- Do not use under occlusive dressings unless specifically instructed by your doctor.
- Inform your doctor if your condition worsens or does not improve after 2 weeks of treatment.
