Description
Overview of the medicine
Vita-D3 1000 IU Tablet contains Cholecalciferol (Vitamin D3), which is essential for calcium absorption and bone health. It helps maintain healthy bones, teeth, and supports immune function.
Uses & Indications
Dosage
Adults
Typically 1000-4000 IU daily, depending on deficiency level and physician's recommendation. For deficiency, higher doses initially may be prescribed.
Elderly
Same as adults, but may require higher doses due to reduced skin synthesis and absorption.
Renal_impairment
Use with caution. In severe renal impairment, active vitamin D metabolites (e.g., calcitriol) may be preferred. Consult a physician.
How to Take
Swallow the tablet whole with water, preferably with a meal to enhance absorption.
Mechanism of Action
Cholecalciferol is converted to its active form, calcitriol, in the body. Calcitriol acts as a hormone to regulate calcium and phosphate homeostasis, promoting their absorption from the gut and deposition in bones.
Pharmacokinetics
Onset
Effects on calcium and phosphate levels typically seen within days to weeks, with full effect on bone mineralization taking longer.
Excretion
Mainly excreted in bile and feces. A small amount is excreted in urine.
Half life
The half-life of calcifediol (25(OH)D3) is about 15-20 days, while calcitriol (1,25(OH)2D3) has a half-life of 4-6 hours. Cholecalciferol itself is rapidly cleared.
Absorption
Readily absorbed from the gastrointestinal tract, especially in the presence of fat. Peak plasma concentrations are reached within 4-5 hours.
Metabolism
Converted in the liver to 25-hydroxyvitamin D (calcifediol), then in the kidneys to the active form, 1,25-dihydroxyvitamin D (calcitriol).
Side Effects
Contraindications
- Hypercalcemia (high blood calcium levels)
- Hypervitaminosis D (Vitamin D toxicity)
- Hypersensitivity to Cholecalciferol or any excipients
Drug Interactions
Corticosteroids
May reduce the effects of Vitamin D.
Thiazide diuretics
May increase risk of hypercalcemia.
Phenytoin, barbiturates
May accelerate metabolism of Vitamin D, reducing its effectiveness.
Orlistat, Cholestyramine, mineral oil
May impair absorption of Vitamin D.
Storage
Store in a cool, dry place, below 30°C. Protect from light and moisture. Keep out of reach of children.
Overdose
Symptoms include hypercalcemia (nausea, vomiting, weakness, polyuria, dehydration), kidney stones, and rarely, kidney failure. Treatment involves discontinuing Vitamin D, low-calcium diet, and hydration. Severe cases may require corticosteroids.
Pregnancy & Lactation
Generally considered safe and necessary during pregnancy and lactation, but doses should be determined by a healthcare professional.

Lab Monitoring
- Serum 25-hydroxyvitamin D levels
- Serum calcium levels
- Serum phosphate levels
- Renal function (creatinine, BUN)

- Educate patients on symptoms of hypercalcemia.
Patient Guidelines
- Take as directed by your doctor.
- Do not exceed the recommended dose.
- Inform your doctor about all other medications and supplements you are taking.
