Uses & Indications
Dosage
Adults
For prevention of Vitamin D deficiency: 2000 IU (1 tablet) daily. For treatment of deficiency, higher doses may be prescribed by a physician based on serum 25(OH)D levels.
Elderly
Similar to adult dosage. Older adults may have reduced skin synthesis of Vitamin D and may require supplementation.
Renal_impairment
Use with caution. High doses may lead to hypercalcemia. Active forms of Vitamin D may be preferred in severe renal failure. Consult a physician.
How to Take
Take the tablet orally, preferably with the largest meal of the day to enhance absorption. Do not chew or crush the tablet, swallow it whole with water.
Mechanism of Action
Cholecalciferol (Vitamin D3) is biologically inactive but is converted in the liver to 25-hydroxycholecalciferol (calcidiol) and then in the kidneys to the active form, 1,25-dihydroxycholecalciferol (calcitriol). Absorption
Cholecalciferol is well absorbed from the gastrointestinal tract, especially in the presence of bile salts and dietary fat. Peak plasma concentrations are reached within 2-4 hours.
Metabolism
Initially hydroxylated in the liver to 25-hydroxycholecalciferol (calcidiol), which is the major circulating form. Further hydroxylated in the kidneys to 1,25-dihydroxycholecalciferol (calcitriol), the biologically active form.
