Mechanism of Action
Salbutamol selectively stimulates beta-2 adrenergic receptors in bronchial smooth muscle, leading to muscle relaxation, bronchodilation, and increased airflow. It also inhibits the release of mast cell mediators.
Pharmacokinetics
Onset
30 minutes (oral)
Excretion
Mainly via urine as metabolites and unchanged drug.
Half life
4-6 hours
Absorption
Rapidly absorbed from the gastrointestinal tract. Oral bioavailability is 50-85% due to first-pass metabolism.
Metabolism
Primarily hepatic, largely to an inactive sulfate conjugate.
Side Effects

- Threatened abortion
Drug Interactions
Diuretics
May cause hypokalemia, increasing risk of arrhythmias.
MAOIs/TCAs
Increased risk of cardiovascular effects.
Beta-blockers
May antagonize the effects of salbutamol, potentially causing severe bronchospasm.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms include tachycardia, tremors, palpitations, hyperglycemia, and hypokalemia. Treatment is supportive and symptomatic; consider a cardioselective beta-blocker with caution for cardiac symptoms.
Pregnancy & Lactation
Use during pregnancy only if the potential benefit outweighs the potential risk to the fetus. Excreted in breast milk, use with caution in breastfeeding mothers.
