Description
Overview of the medicine
Avloclav 250 mg tablet contains amoxicillin, a penicillin antibiotic, and clavulanate potassium, a beta-lactamase inhibitor. It is used to treat bacterial infections that are resistant to amoxicillin alone.
Uses & Indications
Dosage
Adults
Typically, 250 mg/62.5 mg tablet three times daily (every 8 hours) for 7-10 days, depending on the severity and type of infection. Higher doses may be used for severe infections.
Elderly
No dose adjustment is generally required unless there is severe renal impairment. Monitor renal function.
Renal_impairment
Dose adjustment is required for patients with creatinine clearance < 30 mL/min. For example, 250 mg/62.5 mg every 12-24 hours.
How to Take
Administer orally at the start of a meal to minimize potential gastrointestinal intolerance and enhance absorption. Take with a full glass of water.
Mechanism of Action
Amoxicillin inhibits bacterial cell wall synthesis. Clavulanate potassium inactivates beta-lactamase enzymes produced by bacteria, thereby extending amoxicillin's spectrum of activity against beta-lactamase producing strains.
Pharmacokinetics
Onset
Typically within 1 hour.
Excretion
Primarily excreted unchanged by the kidneys, via glomerular filtration and tubular secretion. A small portion is excreted in feces.
Half life
Amoxicillin: 1-1.5 hours; Clavulanate: 1 hour.
Absorption
Rapidly and well absorbed after oral administration, bioavailability is approximately 70% for both components. Food can slightly delay absorption but does not significantly affect total absorption.
Metabolism
Both components are minimally metabolized; Amoxicillin undergoes partial metabolism to inactive metabolites. Clavulanate is more extensively metabolized.
Side Effects
Contraindications
- History of severe hypersensitivity reactions (e.g., anaphylaxis) to beta-lactam antibiotics (e.g., penicillins, cephalosporins)
- History of amoxicillin-clavulanate-associated jaundice or hepatic dysfunction
Drug Interactions
Probenecid
Increases and prolongs plasma levels of amoxicillin (not clavulanate).
Methotrexate
May increase methotrexate toxicity; monitor levels.
Oral contraceptives
May reduce efficacy of oral contraceptives (use alternative contraception).
Anticoagulants (e.g., Warfarin)
May prolong prothrombin time; monitor INR.
Storage
Store in a cool, dry place, below 30°C. Protect from light and moisture. Keep out of reach of children.
Overdose
Symptoms may include gastrointestinal upset, rash, or convulsions (in patients with impaired renal function or high doses). Treatment is supportive; hemodialysis can remove amoxicillin and clavulanate.
Pregnancy & Lactation
Pregnancy Category B. Use during pregnancy only if clearly needed. Excreted in breast milk; use with caution during breastfeeding.
