Avloclav-875 mg Tablet is a combination antibiotic containing amoxicillin and clavulanic acid. Amoxicillin is a penicillin-type antibiotic that fights bacteria, and clavulanic acid helps prevent certain bacteria from becoming resistant to amoxicillin. It is used to treat a wide variety of bacterial infections.
Uses & Indications
Dosage
Adults
One Avloclav-875 mg tablet (875 mg amoxicillin/125 mg clavulanic acid) twice daily, usually for 7-10 days, depending on the severity and type of infection. Doses should be taken at the start of a meal.
Elderly
No dosage adjustment is generally required in elderly patients, but renal function should be monitored.
Renal_impairment
Dosage adjustment is necessary for patients with creatinine clearance less than 30 mL/min. Consult product information for specific recommendations.
How to Take
Take the tablet whole with a glass of water at the start of a meal to reduce potential gastrointestinal upset and enhance absorption. Do not crush or chew. Space doses evenly.
Mechanism of Action
Amoxicillin works by inhibiting bacterial cell wall synthesis, leading to bacterial lysis and death. Clavulanic acid is a beta-lactamase inhibitor that binds irreversibly to and inactivates beta-lactamase enzymes produced by some bacteria. This protects amoxicillin from hydrolysis by these enzymes, allowing it to exert its bactericidal effect.

Bactericidal effects typically begin within 1 hour of administration.
Excretion
Both amoxicillin and clavulanic acid are primarily eliminated via renal excretion (glomerular filtration and tubular secretion).
Half life
Amoxicillin has a plasma half-life of approximately 1 to 1.5 hours. Clavulanic acid has a plasma half-life of approximately 1 hour.
Absorption
Both amoxicillin and clavulanic acid are well absorbed from the gastrointestinal tract after oral administration. Peak plasma concentrations are achieved in approximately 1 to 2.5 hours.
Metabolism
Amoxicillin is minimally metabolized in the body. Clavulanic acid is extensively metabolized to inactive metabolites.
Side Effects
Contraindications
- •History of severe hypersensitivity (e.g., anaphylaxis) to beta-lactam antibiotics (e.g., penicillins, cephalosporins)
- •History of amoxicillin-clavulanate associated jaundice or hepatic dysfunction
- •Infectious mononucleosis (increased risk of erythematous rash)
Drug Interactions
Probenecid
Decreases renal tubular secretion of amoxicillin, leading to increased and prolonged amoxicillin blood levels.
Allopurinol
Increased incidence of skin rash, particularly in patients with hyperuricemia.
Methotrexate
May increase methotrexate toxicity due to reduced renal excretion.
Oral contraceptives
May reduce the efficacy of oral contraceptives; use of alternative birth control methods should be considered.
Warfarin and other oral anticoagulants
Increased prothrombin time (INR) and bleeding risk. Close monitoring of INR is recommended.
Storage
Store below 30°C in a dry place, away from direct sunlight and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose may include gastrointestinal symptoms (nausea, vomiting, diarrhea) and disturbances of fluid and electrolyte balance. Renal impairment can lead to amoxicillin crystalluria. Management involves symptomatic and supportive treatment. Hemodialysis can be used to remove amoxicillin and clavulanic acid from the circulation.
Pregnancy & Lactation
Pregnancy Category B. Studies in animals have not shown harm to the fetus. However, use during pregnancy should only be considered if the potential benefit outweighs the potential risk. Both amoxicillin and clavulanic acid are excreted in breast milk. Generally considered safe for breastfeeding infants, but monitor for diarrhea, candidiasis (thrush), or sensitization in the infant.
