Description
Overview of the medicine
Delight 10000 IU Oral Solution is a vitamin D supplement used to treat and prevent vitamin D deficiency, promote calcium absorption, and support bone health.
Uses & Indications
Dosage
Adults
For severe deficiency: 10000 IU once weekly for 8-12 weeks, then maintenance dose as advised by physician. For maintenance: 10000 IU once every 2-4 weeks or as directed by a physician.
Elderly
Similar to adult dosage, but caution should be exercised and vitamin D levels monitored, especially if renal function is impaired.
Renal_impairment
Use with caution and monitor serum calcium and phosphate levels closely. Dosage adjustment may be required.
How to Take
The oral solution should be taken orally. It can be taken directly or mixed with food or a drink. It is preferably taken with the largest meal of the day to enhance absorption.
Mechanism of Action
Cholecalciferol (Vitamin D3) is converted in the body to calcitriol, an active form of vitamin D. Calcitriol regulates calcium and phosphate homeostasis, promoting their absorption from the gut and regulating bone mineralization.
Pharmacokinetics
Onset
Therapeutic effects may take several days to weeks to manifest, as conversion to active metabolites and subsequent physiological effects are gradual.
Excretion
Excreted primarily in the bile and feces, with only a small amount eliminated via the urine.
Half life
The half-life of 25-hydroxyvitamin D (calcifediol), the main circulating form, is approximately 19-48 hours, but the storage in adipose tissue results in a much longer overall effect duration.
Absorption
Cholecalciferol is readily absorbed from the small intestine, especially when taken with fatty meals. Peak plasma concentrations are reached within 4-5 hours.
Metabolism
Initially hydroxylated in the liver to 25-hydroxyvitamin D (calcifediol), then further hydroxylated in the kidneys to 1,25-dihydroxyvitamin D (calcitriol), the biologically active form.
Side Effects
Contraindications
- •Hypercalcemia (high blood calcium levels)
- •Hypervitaminosis D (excessive vitamin D levels)
- •Nephrolithiasis (kidney stones)
- •Severe renal impairment (unless vitamin D replacement is specifically indicated and monitored)
Drug Interactions
Corticosteroids
May reduce the effects of vitamin D.
Thiazide Diuretics
May increase the risk of hypercalcemia.
Digitalis Glycosides
Hypercalcemia can potentiate the effects of digitalis, leading to cardiac arrhythmias.
Phenytoin, Barbiturates
May accelerate the metabolism of vitamin D, reducing its effect.
Storage
Store below 30°C in a dry place, away from direct sunlight and moisture. Keep out of reach of children. Do not freeze.
Overdose
Symptoms of overdose include hypercalcemia (nausea, vomiting, thirst, polyuria, constipation, muscle weakness), renal impairment, and cardiac arrhythmias. Treatment involves discontinuing the drug, hydration, and managing hypercalcemia.
Pregnancy & Lactation
Safe when used in recommended doses. High doses should be avoided during pregnancy and lactation due to potential risks of hypercalcemia in the fetus or infant. Consult a doctor.
