Uses & Indications
Dosage
Adults
For prevention of Vitamin D deficiency, typically 1000-2000 IU daily. For treatment of mild deficiency, higher doses may be prescribed by a physician.
Elderly
Similar to adult dosage; however, some elderly individuals may require slightly higher doses due to reduced skin synthesis and absorption. Consult a doctor.
Renal_impairment

How to Take

Mechanism of Action
Cholecalciferol (Vitamin D3) is biologically inactive and is converted in the liver to 25-hydroxycholecalciferol (calcifediol) and then in the kidneys to 1,25-dihydroxycholecalciferol (calcitriol), the active form. Calcitriol promotes calcium absorption from the gastrointestinal tract, reabsorption of calcium in the renal tubules, and mobilization of calcium from bone. It also affects phosphate metabolism and has immunomodulatory properties.

Onset
The full therapeutic effect of Vitamin D supplementation may take several weeks to manifest as blood levels slowly increase and stabilize.
Excretion
Mainly excreted in the bile and feces, with only a small amount excreted in urine.
Half life
The elimination half-life of 25-hydroxyvitamin D (calcifediol), the major circulating form, is approximately 2-3 weeks. The active form, 1,25-dihydroxyvitamin D (calcitriol), has a half-life of about 15 hours.
Absorption
Cholecalciferol is readily absorbed from the small intestine, especially in the presence of bile salts and dietary fats.
Metabolism
Initially hydroxylated in the liver by 25-hydroxylase to 25-hydroxycholecalciferol, which is then further hydroxylated in the kidneys by 1-alpha-hydroxylase to the active form, 1,25-dihydroxycholecalciferol.
