Overview of the medicine
Ofloxacin is a synthetic broad-spectrum antibacterial agent belonging to the fluoroquinolone class. It is effective against a wide range of gram-positive and gram-negative bacteria, used to treat various bacterial infections.
Uses & Indications
Dosage
Adults
Typical dosage for adults is 200 mg to 400 mg twice daily, adjusted based on infection type and severity. For suspension, this translates to 5-10 ml twice daily.
Elderly
Dosage adjustment may be necessary for elderly patients, particularly those with impaired renal function.
Renal_impairment
Dosage should be reduced in patients with renal impairment based on creatinine clearance.
How to Take
For oral administration. Shake the suspension well before use. Can be taken with or without food. Ensure adequate hydration during treatment.
Mechanism of Action
Ofloxacin acts by inhibiting bacterial DNA gyrase and topoisomerase IV, essential enzymes involved in DNA replication, transcription, repair, and recombination. This leads to disruption of bacterial DNA and cell death.

Side Effects
Contraindications
- Hypersensitivity to Ofloxacin or any other fluoroquinolone

Drug Interactions
Warfarin
May enhance anticoagulant effects, leading to an increased risk of bleeding. Monitor INR closely.
Cimetidine, Cyclosporine
May alter Ofloxacin metabolism or elimination, requiring dose adjustments.
Nonsteroidal Anti-inflammatory Drugs (NSAIDs)
May increase CNS stimulation and the risk of seizures when co-administered with fluoroquinolones.
Antacids containing magnesium or aluminum, iron, and zinc supplements
Can significantly reduce Ofloxacin absorption. Administer at least 2 hours before or after Ofloxacin.
Storage
Store below 30°C, protected from light and moisture. Do not freeze. Keep out of reach of children.
Overdose
In case of overdose, gastric lavage and supportive care are recommended. Maintain adequate hydration. Symptoms may include CNS disturbances (e.g., confusion, seizures), gastrointestinal upset (e.g., nausea, vomiting).
Pregnancy & Lactation
Pregnancy Category C. Should be used during pregnancy only if the potential benefit justifies the potential risk to the fetus. Ofloxacin is excreted into breast milk and therefore not recommended during lactation.
