Aceflex 100 mg Tablet contains Aceclofenac, a non-steroidal anti-inflammatory drug (NSAID) used for symptomatic treatment of pain and inflammation in conditions like osteoarthritis, rheumatoid arthritis, and ankylosing spondylitis.
Uses & Indications
Dosage
Adults
The recommended dose is 100 mg twice daily, preferably with or after food.
Elderly
The usual adult dosage can be given, but caution is advised. A lower starting dose may be considered.
Renal_impairment
No evidence that the dose of Aceclofenac needs to be modified in patients with mild renal impairment, but caution is advised. Should be avoided in severe renal impairment.
How to Take
Take Aceflex 100 mg Tablet orally, preferably with or after food to minimize gastrointestinal discomfort. Swallow the tablet whole with water; do not chew or crush.
Mechanism of Action
Aceclofenac exerts its anti-inflammatory and analgesic effects primarily by inhibiting the enzyme cyclo-oxygenase (COX), leading to decreased prostaglandin synthesis. It shows some selectivity for COX-2 over COX-1, which may contribute to a better gastrointestinal tolerability profile.

Rapid and complete absorption after oral administration, with peak plasma concentrations reached in 1.25 to 3 hours.
Metabolism
Extensive metabolism in the liver, primarily via CYP2C9, to 4'-hydroxyaceclofenac and other metabolites.
Side Effects
Contraindications
- Hypersensitivity to Aceclofenac or any components of the tablet.
- Patients with active gastrointestinal ulceration, bleeding or perforation.
- Patients who have experienced asthma, acute rhinitis, angioedema, or urticaria after taking aspirin or other NSAIDs.
- Severe heart failure, hepatic failure, or renal failure.
- Third trimester of pregnancy.

Increased risk of gastrointestinal bleeding.

Increased plasma lithium levels.
Warfarin

Methotrexate

Overdose
Symptoms of overdose may include nausea, vomiting, epigastric pain, gastrointestinal bleeding, diarrhea, dizziness, disorientation, excitation, coma, drowsiness, tinnitus, and convulsions. In cases of significant poisoning, acute renal failure and liver damage are possible. Management involves symptomatic and supportive treatment, including gastric lavage, activated charcoal, and monitoring of vital signs and laboratory parameters.
Pregnancy & Lactation
Pregnancy Category C (first and second trimester), Category D (third trimester). Aceclofenac should be avoided during the third trimester of pregnancy due to the risk of premature closure of the ductus arteriosus and potential renal dysfunction in the fetus. Use with caution in the first and second trimesters only if the potential benefit justifies the potential risk to the fetus. Not recommended during lactation as Aceclofenac is excreted in breast milk.
