Overview of the medicine
Golixen 150 mg tablet contains Fluconazole, an antifungal medication used to treat a variety of fungal and yeast infections. It is effective against systemic candidiasis, cryptococcal meningitis, and vaginal candidiasis.
Uses & Indications
Dosage
Adults
For vaginal candidiasis: 150 mg as a single oral dose. For oropharyngeal candidiasis: 50-100 mg once daily for 7-14 days. For systemic candidiasis: 400 mg on day 1, then 200-400 mg once daily. Dosage varies significantly based on infection type and severity.
Elderly
No specific dose adjustment is usually required unless there is renal impairment.
Renal_impairment
Dose adjustment required. For creatinine clearance 21-50 mL/min, half the recommended dose. For creatinine clearance <20 mL/min, 25% of the recommended dose.
How to Take
Golixen tablets can be taken with or without food. Swallow the tablet whole with a glass of water.
Mechanism of Action
Fluconazole selectively inhibits fungal cytochrome P450-dependent 14α-lanosterol demethylation, which is an essential step in fungal ergosterol biosynthesis. This inhibition leads to the accumulation of 14α-methyl sterols and a decrease in ergosterol, causing disruption of the fungal cell membrane and cell death.

Within 1-2 days for most infections; single dose for vaginal candidiasis may show relief within 24 hours.
Excretion
Primarily excreted unchanged in the urine (approximately 80% of the dose).

Well absorbed from the gastrointestinal tract; oral bioavailability is >90%. Peak plasma concentrations occur within 1-2 hours.

Side Effects
Contraindications
- Hypersensitivity to fluconazole, other azole antifungals, or any component of the formulation.
- Co-administration with terfenadine in patients receiving multiple doses of fluconazole 400 mg/day or higher.
- Co-administration with cisapride, astemizole, pimozide, quinidine, or erythromycin.

Warfarin
Increased prothrombin time; monitor INR.
Phenytoin
Increased phenytoin levels; monitor phenytoin concentrations.
Rifampicin
Decreased fluconazole levels; consider increasing fluconazole dose.
Cyclosporine

Storage
