Description
Overview of the medicine
Lamidin 100 mg Tablet contains Lamivudine, an antiviral medication used to treat chronic Hepatitis B virus (HBV) infection. It works by interfering with the replication of the virus.
Uses & Indications
Dosage
Adults
For chronic Hepatitis B: 100 mg once daily.
Elderly
No specific dose adjustment is required in elderly patients, but renal function should be monitored.
Renal_impairment
Dose adjustment is required in patients with creatinine clearance <50 mL/min. For CrCl 30-49 mL/min: 50 mg once daily; for CrCl 15-29 mL/min: 25 mg once daily; for CrCl 5-14 mL/min: 25 mg first dose, then 25 mg every other day; for CrCl <5 mL/min: 25 mg first dose, then 25 mg every 3-7 days.
How to Take
Lamidin 100 mg Tablet should be taken orally, with or without food. It is important to take the medication regularly at the same time each day.
Mechanism of Action
Lamivudine is a nucleoside analogue. It is intracellularly phosphorylated to its active triphosphate form. Lamivudine triphosphate inhibits viral reverse transcriptase by competing with the natural substrate deoxycytidine triphosphate and by incorporation into viral DNA, resulting in DNA chain termination.
Pharmacokinetics
Onset
Within hours for antiviral effect, clinical improvements seen over weeks to months.
Excretion
Mainly excreted unchanged in the urine (approximately 70% of the dose) by glomerular filtration and active tubular secretion.
Half life
Plasma elimination half-life is 5-7 hours.
Absorption
Rapidly absorbed after oral administration; bioavailability is approximately 80-86%. Peak plasma concentrations occur 0.5-1.5 hours post-dose.
Metabolism
Not extensively metabolized. Primarily eliminated unchanged.
Side Effects
Contraindications
- Hypersensitivity to lamivudine or any component of the formulation.
Drug Interactions
Emtricitabine
Should not be used concurrently due to similar mechanism of action and potential for additive toxicities.
Trimethoprim/Sulfamethoxazole
Increases lamivudine plasma concentrations; may require dose adjustment.
Storage
Store below 30°C in a dry place, away from direct light and moisture. Keep out of reach of children.
Overdose
Limited experience with acute overdose. In studies, high doses caused no unexpected adverse effects. Treatment should be symptomatic and supportive.
Pregnancy & Lactation
Pregnancy Category C. Use in pregnancy only if the potential benefit justifies the potential risk to the fetus. It is excreted in breast milk; advise mothers not to breastfeed if being treated with lamivudine for HBV.
