Overview of the medicine
Rebalin 75 mg Capsule contains Pregabalin, an anticonvulsant used to treat neuropathic pain, epilepsy, and generalized anxiety disorder. It works by decreasing abnormal excitement in the brain.
Uses & Indications
Dosage
Adults
Typically initiated at 75 mg twice daily or 50 mg three times daily, which may be increased based on response and tolerability, up to a maximum of 300 mg twice daily or 200 mg three times daily (600 mg/day).
Elderly
Dose adjustments may be necessary due to potential age-related decrease in renal function. Start at a lower dose and titrate cautiously.
Renal_impairment
Dose must be reduced in patients with impaired renal function based on creatinine clearance. For CrCl 30-60 mL/min, 75 mg/day in 2-3 divided doses. For CrCl 15-30 mL/min, 25-50 mg/day in 1-2 divided doses.
How to Take
Take orally with a glass of water, with or without food. Swallow the capsule whole; do not crush, chew, or open.
Mechanism of Action
Pregabalin binds with high affinity to the alpha2-delta (α2-δ) subunit of voltage-gated calcium channels in the central nervous system. This binding is thought to reduce the release of several neurotransmitters, including glutamate, norepinephrine, and substance P, which are involved in pain transmission and seizure activity.
Pharmacokinetics
Onset
Analgesic effects typically observed within 1 week for neuropathic pain. Anxiolytic effects may appear within days to weeks.
Excretion
Primarily excreted unchanged in the urine (approximately 98% of the dose is excreted as unchanged drug).
Half life
Approximately 6.3 hours
Absorption
Rapidly and well absorbed after oral administration, with peak plasma concentrations occurring within 1.5 hours. Oral bioavailability is >90% and is not affected by food.
Metabolism
Negligible; Pregabalin undergoes negligible metabolism in humans. Less than 0.1% of the dose is recovered as metabolites in the urine.

Contraindications
- Known hypersensitivity to pregabalin or any component of the formulation
Drug Interactions
Angiotensin-Converting Enzyme (ACE) Inhibitors
May lead to increased risk of peripheral edema, though the mechanism is not fully understood.
CNS Depressants (e.g., alcohol, benzodiazepines, opioids)
Concomitant use may potentiate CNS depression (e.g., sedation, dizziness, somnolence, decreased alertness).
Storage
Store in a cool, dry place below 30°C, protected from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose include somnolence, confusion, agitation, and restlessness. Treatment is supportive and symptomatic, and hemodialysis may be considered in severe cases as pregabalin is dialyzable.
Pregnancy & Lactation
Pregnancy Category C. Should be used during pregnancy only if the potential benefit justifies the potential risk to the fetus. Not recommended for use during lactation as pregabalin is excreted in human milk.
