Description
Overview of the medicine
A-B1 100 mg Tablet contains Thiamine Hydrochloride, also known as Vitamin B1. It is an essential water-soluble vitamin required for various metabolic processes, nerve function, and heart health. Used to treat and prevent thiamine deficiency.
Uses & Indications
Dosage
Adults
For mild deficiency: 10-25 mg daily. For severe deficiency/Beriberi: 100 mg daily or several times daily, as advised by a physician.
Elderly
Same as adult dosage, adjust based on clinical condition.
Renal_impairment
No specific dosage adjustment required, as it is a water-soluble vitamin.
How to Take
Oral administration. Tablets can be taken with or without food.
Mechanism of Action
Thiamine is converted into its active form, thiamine pyrophosphate (TPP), which acts as a coenzyme in carbohydrate metabolism, specifically in the decarboxylation of alpha-keto acids and transketolase reactions.
Pharmacokinetics
Onset
Therapeutic effects may be observed within hours to days for deficiency symptoms.
Excretion
Excreted in the urine primarily as metabolites or unchanged thiamine, especially after high doses.
Half life
Approximately 98 minutes (active metabolite).
Absorption
Readily absorbed from the gastrointestinal tract, primarily in the duodenum, through active transport at low concentrations and passive diffusion at high concentrations. Absorption is dose-dependent and saturable.
Metabolism
Metabolized in the liver to thiamine pyrophosphate (TPP), its active form, and other inactive metabolites.
Side Effects
Contraindications
- Hypersensitivity to thiamine or any component of the formulation
Drug Interactions
Alcohol
Chronic alcohol consumption may impair thiamine absorption and utilization.
Diuretics (loop diuretics)
May increase urinary excretion of thiamine, potentially leading to deficiency.
Storage
Store below 30°C in a dry place, away from direct sunlight and moisture. Keep out of reach of children.
Overdose
Thiamine has a wide therapeutic index, and overdose is rare with oral administration. Extremely high doses may cause mild gastrointestinal upset or allergic reactions in hypersensitive individuals. Treatment is supportive.
Pregnancy & Lactation
Generally considered safe and essential during pregnancy and lactation. Consult a physician for appropriate dosing.

Lab Monitoring
- No routine lab monitoring is required for oral thiamine supplementation. In severe deficiency, thiamine levels or erythrocyte transketolase activity may be monitored.
Doctor Notes
- Counsel patients on dietary sources of thiamine.
- Monitor for signs of deficiency in high-risk groups (e.g., alcoholics, patients with malabsorption).
Patient Guidelines
- Take as directed by your doctor or pharmacist.
- Do not exceed the recommended dose.
- Store in a cool, dry place away from light and moisture.
- If symptoms of deficiency persist, consult your doctor.
Missed Dose Advice
If a dose is missed, take it as soon as you remember. If it is almost time for your next dose, skip the missed dose and resume your regular dosing schedule. Do not double the dose.
Driving Precautions
Thiamine is not known to impair the ability to drive or operate machinery.
Lifestyle Advice
- Maintain a balanced diet rich in thiamine-containing foods (whole grains, nuts, seeds, pork).
- Limit alcohol consumption, as it can impair thiamine absorption.
