Overview of the medicine
Alargol 10 mg Tablet contains Loratadine, a non-drowsy antihistamine used to relieve symptoms associated with allergic rhinitis (hay fever) and chronic urticaria (hives). It helps in reducing sneezing, runny nose, itchy or watery eyes, and skin itching.
Uses & Indications
Dosage
Adults
10 mg once daily.
Elderly
No dosage adjustment is generally necessary in elderly patients with normal renal and hepatic function.
Renal_impairment
For patients with severe renal impairment (creatinine clearance < 30 mL/min), initial dose should be 10 mg every other day.
How to Take
Alargol tablet can be taken with or without food. Swallow the tablet whole with water.
Mechanism of Action
Loratadine is a tricyclic antihistamine with selective peripheral H1-receptor antagonist activity. It prevents histamine from binding to H1-receptors, thereby inhibiting the symptoms of allergy, without significant central nervous system effects.

Within 1-3 hours.
Excretion
Approximately 40% excreted in urine and 42% in feces over 10 days, primarily in the form of conjugated metabolites.
Half life
Loratadine: 8.4 hours (range 3-20 hours). Descarboethoxyloratadine (active metabolite): 28 hours (range 8.8-92 hours).
Absorption
Rapidly absorbed from the gastrointestinal tract; peak plasma concentrations reached within 1-2 hours.
Metabolism
Extensively metabolized in the liver, primarily by CYP3A4 and CYP2D6, to its major active metabolite, descarboethoxyloratadine.
Side Effects
Contraindications
- Hypersensitivity to Loratadine or any component of the formulation.
- Known allergy to descarboethoxyloratadine.
Drug Interactions
Ketoconazole, Erythromycin, Cimetidine
May increase plasma concentrations of Loratadine and its active metabolite, though no clinically significant adverse effects have been reported.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose include somnolence, tachycardia, and headache. In the event of an overdose, symptomatic and supportive treatment is recommended. Gastric lavage and activated charcoal may be considered.
Pregnancy & Lactation
Pregnancy Category B. Use during pregnancy only if clearly needed. Loratadine and its metabolite pass into breast milk; therefore, caution should be exercised when administered to a nursing mother.
