Amoxicillin is a broad-spectrum penicillin antibiotic used to treat a variety of bacterial infections.
Uses & Indications
Dosage
Adults
Typical adult dose is 250 mg to 500 mg every 8 hours, or 500 mg to 875 mg every 12 hours, depending on the severity and type of infection.
Elderly
Similar to adults, but dose adjustment may be necessary in case of impaired renal function.
Renal_impairment
Dose adjustment required based on creatinine clearance. For CrCl <30 mL/min, dosage intervals should be extended.
How to Take
Take orally with a glass of water, with or without food. Complete the full course as prescribed.
Mechanism of Action
It works by inhibiting bacterial cell wall synthesis, leading to bacterial cell lysis and death.

Excretion
Primarily excreted unchanged in the urine by glomerular filtration and tubular secretion.

Contraindications

Drug Interactions
Warfarin
May increase anticoagulant effect, requiring INR monitoring and dose adjustment.
Allopurinol
Increased incidence of rash when co-administered.
Methotrexate
May decrease methotrexate clearance, leading to increased toxicity.
Oral contraceptives
May reduce efficacy of oral contraceptives (rare, but advise caution).
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose include gastrointestinal disturbances (nausea, vomiting, diarrhea) and fluid-electrolyte imbalance. In very high doses or in patients with impaired renal function, crystalluria and renal impairment may occur. Treatment is symptomatic and supportive; hemodialysis may be used to remove Amoxicillin from circulation.
Pregnancy & Lactation
Pregnancy Category B. Generally considered safe during pregnancy. Excreted in small amounts in breast milk; generally compatible with breastfeeding.
