Anadol 100 mg Injection contains Tramadol Hydrochloride, which is a synthetic opioid analgesic used for the relief of moderate to severe pain. It acts on opioid receptors in the brain and spinal cord to reduce the perception of pain. It is often used in hospital settings for post-operative pain or acute severe pain.
Uses & Indications
Dosage
Adults
Typically, 50-100 mg IV or IM every 4-6 hours as needed, not to exceed 400 mg/day.
Elderly
No specific dose adjustment needed unless severe renal or hepatic impairment. Doses may be reduced for patients over 75 years.
Renal_impairment
Reduce dose and/or increase dosing interval in patients with creatinine clearance less than 30 mL/min (e.g., 50-100 mg every 12 hours).
How to Take
Administer slowly intravenously over 2-3 minutes or intramuscularly. Ensure proper aseptic technique.
Mechanism of Action
Tramadol is a centrally acting analgesic. It binds to μ-opioid receptors in the central nervous system. Additionally, it inhibits the reuptake of norepinephrine and serotonin, which contributes to its analgesic effect.

Side Effects
Contraindications
- Hypersensitivity to tramadol or any component of the formulation
- Acute intoxication with alcohol, hypnotics, centrally acting analgesics, opioids, or psychotropic drugs
- Concomitant use with MAO inhibitors or within 14 days of discontinuing them
- Epilepsy not controlled by treatment
Drug Interactions
Quinidine
Increases tramadol levels by inhibiting CYP2D6 metabolism.
Carbamazepine
Significantly decreases tramadol plasma concentrations, reducing analgesic effect.
MAO Inhibitors
Increased risk of seizures and serotonin syndrome.
SSRIs/SNRIs/TCAs
Increased risk of serotonin syndrome and seizures.
CNS Depressants (e.g., benzodiazepines, alcohol)
Additive CNS depression, increased risk of respiratory depression, profound sedation, coma, and death.

Symptoms of overdose include respiratory depression, somnolence, coma, seizures, skeletal muscle flaccidity, cold and clammy skin, constricted pupils, and possibly death. Treatment involves maintaining a patent airway, providing ventilatory support, and administering opioid antagonists like naloxone.
Pregnancy & Lactation
Pregnancy Category C. Not recommended for use during pregnancy, especially near term, due to potential for neonatal opioid withdrawal syndrome. Tramadol is excreted in breast milk; therefore, it is not recommended during breastfeeding.
