Description
Overview of the medicine
Aromatin 1 mg Tablet contains Anastrozole, an aromatase inhibitor used in the treatment of breast cancer in postmenopausal women. It helps to slow or stop the growth of cancer by reducing the body's production of estrogen, which can fuel the growth of certain types of breast cancer.
Uses & Indications
Dosage
Adults
For postmenopausal women, 1 mg orally once daily.
Elderly
No dosage adjustment is generally required for elderly patients.
Renal_impairment
No dosage adjustment is required for mild to moderate renal impairment. Use with caution in patients with severe renal impairment (creatinine clearance < 20 mL/min).
Hepatic_impairment
No dosage adjustment is required for patients with mild hepatic impairment. Use with caution in patients with moderate to severe hepatic impairment.
How to Take
Aromatin 1 mg tablet should be taken orally once daily. It can be taken with or without food. Try to take it at the same time each day to maintain consistent levels in your body.
Mechanism of Action
Anastrozole works by selectively inhibiting the enzyme aromatase, which is responsible for converting androgens into estrogens in peripheral tissues. By reducing the overall estrogen levels in the body, particularly in postmenopausal women, Anastrozole helps to reduce the growth of hormone-receptor-positive breast cancer cells that depend on estrogen for proliferation.
Pharmacokinetics
Onset
Significant estrogen suppression is observed within 24 hours of a single dose, with maximal suppression achieved within 7 days.
Excretion
Approximately 10% is excreted unchanged in urine; about 60% of metabolites are excreted in urine and 11% in feces within 72 hours.
Half life
Approximately 40-50 hours.
Absorption
Rapidly absorbed from the gastrointestinal tract; peak plasma concentrations are usually attained within 2-3 hours.
Metabolism
Extensively metabolized in the liver, primarily via N-dealkylation, hydroxylation, and glucuronidation. The primary metabolite, triazole, is pharmacologically inactive.
Side Effects
Contraindications
- Pre-menopausal women.
- Pregnancy and lactation.
- Known hypersensitivity to anastrozole or any of its excipients.
Drug Interactions
Tamoxifen or estrogen-containing therapies
Concomitant use with tamoxifen or estrogen-containing therapies (e.g., hormone replacement therapy) should be avoided as these may diminish the pharmacological action of Anastrozole.
Storage
Store below 30°C in a dry place, away from direct sunlight and moisture. Keep out of reach of children.
Overdose
There is no specific antidote for anastrozole overdose. Treatment should be symptomatic and supportive. Standard general supportive care, including frequent monitoring of vital signs and close observation of the patient, is indicated.
Pregnancy & Lactation
Anastrozole is contraindicated during pregnancy and lactation. It can cause fetal harm when administered to a pregnant woman. It is not known whether anastrozole is excreted in human milk.

Clinical Trials
Anastrozole has been extensively studied in numerous large-scale clinical trials (e.g., ATAC trial) demonstrating its efficacy and safety in various settings of hormone receptor-positive breast cancer treatment.
Lab Monitoring
- Bone mineral density (BMD) assessments due to increased risk of osteoporosis.
- Lipid profiles (total cholesterol, LDL, HDL) due to potential for increased cholesterol levels.
- Liver function tests (baseline and periodically, especially in patients with hepatic impairment).
Doctor Notes
- Anastrozole is a first-line agent for adjuvant and metastatic hormone-receptor-positive breast cancer in postmenopausal women.
- Monitor bone mineral density (BMD) at baseline and periodically due to the risk of osteoporosis.
- Counsel patients on potential side effects, particularly hot flashes, arthralgia, and vaginal dryness.
- Avoid concomitant use with tamoxifen or estrogen-containing products.
Patient Guidelines
- Take your medication at the same time each day as prescribed by your doctor.
- Do not stop taking Aromatin without consulting your doctor, even if you feel better.
- Report any unusual or severe side effects to your healthcare provider immediately.
- Discuss bone health with your doctor, as this medication can increase the risk of osteoporosis. Ensure adequate calcium and vitamin D intake.
Missed Dose Advice
If you miss a dose, take it as soon as you remember. If it is almost time for your next dose, skip the missed dose and return to your regular dosing schedule. Do not take a double dose to make up for a missed one.
Driving Precautions
Anastrozole may cause somnolence (drowsiness) or dizziness in some patients. Patients should be advised to exercise caution when driving or operating machinery if these symptoms occur.
Lifestyle Advice
- Engage in regular weight-bearing exercises to help maintain bone density.
- Maintain a balanced diet rich in calcium and vitamin D.
- Avoid smoking and excessive alcohol consumption, as these can negatively affect bone health.
