Bestcol 10 mg Tablet contains Rosuvastatin, a statin medication used to lower high cholesterol and triglycerides, and to reduce the risk of heart attack, stroke, and other heart-related complications. It works by reducing the amount of cholesterol made by the liver.
Uses & Indications
Dosage
Adults
Initial dose is usually 10 mg once daily. Dosage may be adjusted at 2-4 week intervals up to a maximum of 40 mg once daily, based on lipid levels and patient response.
Elderly
No specific dose adjustment is needed for patients >70 years old; however, a starting dose of 5 mg may be considered.
Renal_impairment
For patients with severe renal impairment (CrCl < 30 mL/min), a starting dose of 5 mg once daily is recommended, and the dose should not exceed 20 mg once daily.
How to Take
Take Bestcol 10 mg Tablet orally, once daily, with or without food. It is generally recommended to take it at approximately the same time each day.
Mechanism of Action
Rosuvastatin competitively inhibits HMG-CoA reductase, an enzyme that catalyzes the early, rate-limiting step in cholesterol biosynthesis. This leads to a reduction in hepatic cholesterol synthesis and an upregulation of LDL receptors on the cell surface, which increases LDL-C uptake and catabolism.

Onset
Significant lipid-lowering effects are generally seen within 1 week, with maximal effect usually achieved by 2-4 weeks of therapy.
Excretion
Approximately 90% of rosuvastatin is excreted unchanged in the feces, with a smaller portion excreted in the urine.

The elimination half-life of rosuvastatin is approximately 19 hours.
Absorption
Peak plasma concentrations are achieved approximately 3-5 hours after oral administration. Absolute bioavailability is approximately 20%.
Metabolism
Rosuvastatin undergoes limited metabolism, primarily by CYP2C9 (approximately 10%). The primary metabolite is N-desmethyl rosuvastatin.
