Description
Overview of the medicine
Delot 5 mg tablet contains Desloratadine, a non-drowsy antihistamine used to relieve symptoms of allergies such as sneezing, runny nose, itchy eyes, and hives. It is effective for both seasonal and perennial allergic rhinitis and chronic idiopathic urticaria.
Uses & Indications
Dosage
Adults
5 mg once daily, with or without food.
Elderly
No dosage adjustment generally required, but caution due to potential decreased renal/hepatic function. Consult a doctor.
Renal_impairment
For patients with severe renal impairment (creatinine clearance < 30 mL/min), a starting dose of 5 mg every other day is recommended.
How to Take
Take orally, once daily, with or without food. Swallow the tablet whole with water. Do not crush or chew.
Mechanism of Action
Desloratadine is a potent, non-sedating, long-acting, selective peripheral histamine H1-receptor antagonist. It inhibits histamine release from mast cells and basophils, and also inhibits the release of various inflammatory mediators involved in allergic reactions.
Pharmacokinetics
Onset
Within 1-3 hours.
Excretion
Excreted in urine and feces primarily as metabolites (approximately 87% within 10 days).
Half life
Approximately 27 hours.
Absorption
Rapidly absorbed after oral administration, peak plasma concentration reached in approximately 3 hours.
Metabolism
Undergoes extensive metabolism to 3-hydroxydesloratadine, an active metabolite, primarily via cytochrome P450 enzymes (e.g., CYP2D6 in some individuals).
Side Effects
Contraindications
- Hypersensitivity to desloratadine, loratadine, or any component of the formulation.
Drug Interactions
Cimetidine
Increased plasma concentrations of desloratadine and its active metabolite (not clinically significant).
Fluoxetine
Increased plasma concentrations of desloratadine and its active metabolite (not clinically significant).
Azithromycin
No clinically relevant interaction observed.
Erythromycin
Increased plasma concentrations of desloratadine and its active metabolite (not clinically significant).
Ketoconazole
Increased plasma concentrations of desloratadine and its active metabolite (not clinically significant).
Storage
Store below 30°C in a dry place, away from direct sunlight and moisture. Keep out of reach of children.
Overdose
In case of overdose, symptoms may include drowsiness, tachycardia, and headache. Treatment should be symptomatic and supportive. Desloratadine is not eliminated by hemodialysis.
Pregnancy & Lactation
Pregnancy Category C. Use only if potential benefit justifies the potential risk to the fetus. Desloratadine is excreted into breast milk; therefore, a decision should be made whether to discontinue nursing or to discontinue the drug, taking into account the importance of the drug to the mother.

Clinical Trials
Extensive clinical trials have demonstrated the efficacy and safety of desloratadine for the treatment of allergic rhinitis and chronic idiopathic urticaria. Studies have consistently shown its rapid onset of action and sustained symptom relief with a favorable non-sedating profile.
Lab Monitoring
- No specific laboratory monitoring is generally required. Liver function tests may be considered in patients with pre-existing hepatic impairment or if symptoms suggest liver dysfunction.
Doctor Notes
- Advise patients to take consistently for best results, ideally at the same time each day.
- Counsel on potential mild side effects such as dry mouth or headache, which are usually transient.
- Emphasize the non-sedating nature, but warn about individual variability and to observe their reaction before engaging in activities requiring mental alertness.
Patient Guidelines
- Take the tablet at the same time each day.
- Do not exceed the recommended dose.
- Inform your doctor about all other medications you are taking, including over-the-counter drugs and herbal supplements.
Missed Dose Advice
If you miss a dose, take it as soon as you remember. If it is almost time for your next dose, skip the missed dose and continue with your regular dosing schedule. Do not take a double dose to make up for a missed one.
Driving Precautions
Although Delot is generally non-sedating, individual responses can vary. Patients should be advised to see how they react to the medicine before driving or operating machinery.
Lifestyle Advice
- Avoid known allergens that trigger your symptoms.
- Maintain good hydration.
- Avoid alcohol consumption as it may increase drowsiness, although desloratadine is non-sedating, individual responses vary.
