Description
Overview of the medicine
Ketorin 10 mg Tablet is a non-steroidal anti-inflammatory drug (NSAID) primarily used for the short-term management of moderate to severe acute pain. It works by reducing hormones that cause inflammation and pain in the body.
Uses & Indications
Dosage
Adults
10 mg every 4-6 hours as needed, not to exceed 40 mg per day. Treatment duration should not exceed 5 days.
Elderly
Reduced dosage may be necessary due to potential for impaired renal function. Max 20 mg/day.
Renal_impairment
Contraindicated in advanced renal impairment. For mild impairment, dosage reduction is required; max 10 mg/day.
How to Take
For oral use. Can be taken with food or milk to minimize gastrointestinal upset. Swallow the tablet whole with a glass of water.
Mechanism of Action
Ketorolac inhibits prostaglandin synthesis by blocking cyclooxygenase (COX-1 and COX-2) enzymes. This reduces the formation of prostaglandins, which are mediators of pain, inflammation, and fever.
Pharmacokinetics
Onset
Analgesic effect typically begins within 30-60 minutes.
Excretion
Primarily excreted in the urine (about 91%) and feces (about 6%).
Half life
Approximately 5-6 hours in healthy adults.
Absorption
Rapidly and completely absorbed orally. Peak plasma concentrations are reached within 30-60 minutes after oral administration.
Metabolism
Extensively metabolized in the liver, primarily via glucuronidation.
Side Effects
Contraindications
- Hypersensitivity to ketorolac or other NSAIDs
- Active peptic ulcer, GI bleeding or perforation
- Advanced renal impairment
- Coagulopathy or patients at high risk of bleeding
- As prophylactic analgesic before major surgery
- During labor and delivery
- Children under 16 years
Drug Interactions
Lithium
Increased lithium plasma levels, potentially leading to toxicity.
Methotrexate
Increased methotrexate toxicity.
ACE Inhibitors/ARBs
Increased risk of renal impairment, especially in elderly or volume-depleted patients.
Other NSAIDs/Aspirin
Increased risk of GI adverse effects.
Diuretics (e.g., Furosemide)
Reduced natriuretic effect, potential for renal impairment.
Anticoagulants (e.g., Warfarin)
Increased risk of bleeding.
Storage
Store in a cool, dry place, below 30°C. Protect from light and moisture. Keep out of reach of children.
Overdose
Symptoms may include lethargy, drowsiness, nausea, vomiting, epigastric pain, and gastrointestinal bleeding. Management involves symptomatic and supportive care. Gastric lavage or activated charcoal may be considered if recent ingestion.
Pregnancy & Lactation
Not recommended during pregnancy, especially in the third trimester due to risk of premature closure of the fetal ductus arteriosus. Use with caution during lactation; small amounts may pass into breast milk.

Lab Monitoring
- Renal function (BUN, creatinine)
- Liver function (ALT, AST)
- Complete blood count (CBC) if long-term use (not recommended for ketorolac)
- Stool for occult blood if GI symptoms occur
Doctor Notes
- Emphasize short-term use (max 5 days) due to significant GI and CV risks.
- Assess renal function prior to and during treatment, especially in elderly or compromised patients.
- Concomitant use with other NSAIDs or anticoagulants is generally contraindicated.
- Educate patients on symptoms of GI bleeding and stroke/MI.
