Lansoprazole is a proton pump inhibitor (PPI) used to treat and prevent stomach and esophageal problems like acid reflux, ulcers, and erosive esophagitis. It works by decreasing the amount of acid your stomach makes.
Uses & Indications
Dosage
Adults
Typical dose is 30 mg once daily, usually for 4-8 weeks, depending on the condition. For severe erosive esophagitis or Zollinger-Ellison Syndrome, higher doses may be required.
Elderly
No specific dose adjustment is generally needed for the elderly, unless there is severe hepatic impairment.
Renal_impairment
No dose adjustment is necessary for patients with renal impairment.
Hepatic_impairment
Consider dose reduction in patients with severe hepatic impairment.
How to Take
Take orally, preferably 30 minutes before a meal (e.g., breakfast). Swallow the capsule whole with water; do not crush, chew, or open the capsule.
Mechanism of Action
Lansoprazole irreversibly binds to and inactivates the H+/K+-ATPase proton pump located on the secretory surface of the gastric parietal cells, thereby suppressing gastric acid secretion.

Acid-suppressing effect begins within 1-2 hours.
Excretion
Approximately one-third of the administered dose is excreted in the urine, and two-thirds in the feces as metabolites.
Half life
Plasma elimination half-life is approximately 1.5 hours, but the duration of acid suppression is much longer.

Rapidly absorbed from the gastrointestinal tract following oral administration. Bioavailability is approximately 80-90%.
Metabolism
Extensively metabolized in the liver, primarily by the cytochrome P450 enzymes CYP2C19 and CYP3A4.
Side Effects
Contraindications
