Description
Overview of the medicine
Apoxy 125 mg Suspension is an antibiotic from the penicillin group, used to treat various bacterial infections. It works by stopping the growth of bacteria. It is commonly prescribed for infections of the ear, nose, throat, respiratory tract, skin, and urinary tract.
Uses & Indications
Dosage
Adults
Not typically used in adults at this strength; higher strengths or different forms are used. If used, dosage would be individualized.
Elderly
Dosage adjustment may be necessary in elderly patients with impaired renal function.
Renal_impairment
Dosage reduction or extended dosing interval required based on creatinine clearance.
How to Take
Shake the bottle well before each use. Administer orally, with or without food. Complete the full course of medication as prescribed, even if symptoms improve.
Mechanism of Action
Amoxicillin, a beta-lactam antibiotic, inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins (PBPs). This disrupts the peptidoglycan layer, leading to osmotic lysis and bacterial cell death.
Pharmacokinetics
Onset
Within 30 minutes to 1 hour (for symptomatic relief, full antimicrobial effect takes longer)
Excretion
Mainly excreted unchanged in the urine by glomerular filtration and tubular secretion. About 60-80% of a dose is excreted within 6 hours.
Half life
Approximately 1-1.5 hours in adults with normal renal function.
Absorption
Rapidly and well absorbed from the gastrointestinal tract, about 75-90%. Peak plasma concentrations occur within 1-2 hours.
Metabolism
Limited metabolism, approximately 10-25% is metabolized to inactive penicilloic acid.
Side Effects
Contraindications
- Hypersensitivity to amoxicillin, any other penicillin, or any component of the formulation.
- History of a severe hypersensitivity reaction to another beta-lactam antibiotic (e.g., cephalosporins, carbapenems).
Drug Interactions
Warfarin
May increase the anticoagulant effect of warfarin. Monitor INR.
Methotrexate
May increase methotrexate toxicity due to reduced renal excretion of methotrexate.
Oral contraceptives
May reduce the efficacy of oral contraceptives. Advise use of alternative contraception.
Storage
Store dry powder at room temperature (below 30°C) in a dry place, away from light. Once reconstituted, store in the refrigerator (2-8°C) and discard after specified period.
Overdose
Symptoms of overdose may include gastrointestinal symptoms (nausea, vomiting, diarrhea) and disturbances in fluid and electrolyte balance. In rare cases, crystalluria may be observed leading to renal failure. Treatment is largely symptomatic and supportive. Hemodialysis may be used to remove amoxicillin from circulation.
Pregnancy & Lactation
Amoxicillin is generally considered safe during pregnancy (Category B) and lactation. Consult a doctor before use.
