Aroflo 50 mcg Inhalation Capsule contains salmeterol, a long-acting beta2-adrenergic agonist (LABA), used for the maintenance treatment of asthma and chronic obstructive pulmonary disease (COPD). It helps to relax the muscles around the airways, making them open and easier to breathe.
Uses & Indications
Dosage
Adults
One 50 mcg capsule inhaled twice daily (morning and evening), approximately 12 hours apart.
Elderly
No dosage adjustment required.

How to Take
For oral inhalation only. Do not swallow the capsule. Administer using a suitable dry powder inhaler device. Follow the instructions provided with the inhaler device carefully. It should be used regularly, even when symptom-free, for full benefit.
Mechanism of Action
Salmeterol is a selective, long-acting beta2-adrenergic receptor agonist (LABA). It acts locally in the lungs to relax bronchial smooth muscle by stimulating beta2 receptors, leading to bronchodilation. Its long duration of action is due to its lipophilicity, allowing it to bind to an 'exo-site' on the receptor and continuously stimulate it.

Side Effects
Contraindications
- Primary treatment of status asthmaticus or acute episodes of asthma or COPD requiring intensive measures
- Hypersensitivity to salmeterol or any component of the formulation
Drug Interactions
Diuretics
ECG changes and/or hypokalemia may result from concomitant administration. Use with caution.
Beta-blockers
May block the pulmonary effect of salmeterol and produce severe bronchospasm in asthmatic patients. Should not be used concomitantly.
Tricyclic antidepressants & MAOIs
May potentiate the effect of salmeterol on the cardiovascular system. Use with extreme caution.
CYP3A4 inhibitors (e.g., Ketoconazole, Ritonavir)
May increase salmeterol systemic exposure, leading to increased risk of cardiovascular adverse effects. Use with caution or avoid.
Storage
Store below 30°C in a dry place, away from direct light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose include tremor, palpitations, tachycardia, headache, and hypokalemia. Treatment is symptomatic and supportive, including discontinuation of the drug and appropriate monitoring.

Pregnancy Category C. Use during pregnancy only if the potential benefit justifies the potential risk to the fetus. It is unknown whether salmeterol is excreted in human milk. Exercise caution when administered to a nursing mother.
