Description
Overview of the medicine
Buflex 750 mg tablet contains Naproxen, a non-steroidal anti-inflammatory drug (NSAID) used to relieve pain, inflammation, and stiffness caused by various conditions like arthritis, muscle aches, and menstrual cramps.
Uses & Indications
Dosage
Adults
For chronic conditions: 500-1000 mg daily in one or two doses. For acute conditions (e.g., acute gout): 750 mg initially, then 250 mg every 8 hours until attack subsides. Max 1250 mg on the first day.
Elderly
Use with caution; consider lower doses due to increased risk of side effects, especially GI bleeding and renal impairment.
Renal_impairment
Use with caution; dose reduction may be necessary, and close monitoring of renal function is advised. Avoid in severe renal impairment.
How to Take
Take with food or milk to reduce gastrointestinal upset. Swallow the tablet whole with a full glass of water. Do not crush or chew.
Mechanism of Action
Naproxen inhibits cyclooxygenase (COX-1 and COX-2) enzymes, which are responsible for the synthesis of prostaglandins, mediators of pain and inflammation.
Pharmacokinetics
Onset
1-2 hours for pain relief, longer for chronic inflammatory conditions.
Excretion
Mainly excreted in the urine (about 95%) as metabolites and conjugates, with a small amount in feces.
Half life
Approximately 12-17 hours.
Absorption
Rapidly and completely absorbed from the gastrointestinal tract.
Metabolism
Primarily metabolized in the liver by demethylation and glucuronidation.
Side Effects
Contraindications
- Known hypersensitivity to naproxen or other NSAIDs (e.g., aspirin-induced asthma)
- Active gastrointestinal bleeding or peptic ulcer
- Severe heart failure
- Severe renal impairment
- Severe hepatic impairment
- Third trimester of pregnancy
- Coronary artery bypass graft (CABG) surgery (peri-operative pain)
Drug Interactions
Lithium
Increased lithium plasma levels and toxicity.
Methotrexate
Increased methotrexate toxicity.
ACE inhibitors/ARBs
Reduced antihypertensive effect; increased risk of renal impairment.
Other NSAIDs/Aspirin
Increased risk of GI side effects and bleeding.
Diuretics (e.g., Furosemide)
Reduced diuretic and antihypertensive effects; increased risk of renal impairment.
Anticoagulants (e.g., Warfarin)
Increased risk of bleeding.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms may include drowsiness, heartburn, nausea, vomiting, epigastric pain. More severe cases may involve gastrointestinal bleeding, hypertension, acute renal failure, respiratory depression, and coma. Treatment is symptomatic and supportive; gastric lavage, activated charcoal, and monitoring of vital signs may be required.
Pregnancy & Lactation
Pregnancy: Avoid in the third trimester due to potential premature closure of ductus arteriosus and fetal renal dysfunction. Use in first two trimesters only if clearly needed and benefits outweigh risks. Lactation: Naproxen passes into breast milk in small amounts; use with caution or consider an alternative, especially with long-term use.
