Description
Overview of the medicine
Clindax-B 1%/5% Topical Gel is a combination medication containing an antibiotic (Clindamycin) and a keratolytic agent (Benzoyl Peroxide) used for the topical treatment of acne vulgaris. It works by reducing acne-causing bacteria and unclogging pores.
Uses & Indications
Dosage
Adults
Apply a thin layer to the affected areas once daily, or as directed by a physician, after washing and drying the skin.
Elderly
No specific dosage adjustment required; use as per adult dosage.
Renal_impairment
Not applicable for topical use due to minimal systemic absorption.
How to Take
For topical use only. Wash the affected skin gently, pat dry, and apply a thin layer of gel to the entire affected area (not just individual pimples) once daily in the evening, or as directed by the doctor. Avoid contact with eyes, lips, mouth, and mucous membranes.
Mechanism of Action
Clindamycin is an antibiotic that inhibits bacterial protein synthesis by binding to the 50S ribosomal subunit, thereby suppressing the growth of Propionibacterium acnes (now Cutibacterium acnes). Benzoyl Peroxide has bactericidal activity against C. acnes and also acts as a keratolytic agent, reducing follicular hyperkeratosis and promoting the shedding of dead skin cells.
Pharmacokinetics
Onset
Visible improvement typically within 2-5 weeks of consistent use.
Excretion
Primarily through sebaceous glands and skin surface. Systemically absorbed components are excreted via urine and feces.
Half life
Not well-defined for topical application due to minimal systemic absorption; systemic clindamycin half-life is about 2.4 hours.
Absorption
Minimal systemic absorption after topical application. Clindamycin absorption is approximately 4-5%, while Benzoyl Peroxide is metabolized to benzoic acid, with minimal systemic absorption (about 5%).
Metabolism
Topically applied clindamycin is minimally metabolized. Benzoyl Peroxide is metabolized to benzoic acid in the skin.
Side Effects
Contraindications
- Hypersensitivity to clindamycin, lincomycin, benzoyl peroxide, or any component of the formulation.
- History of regional enteritis, ulcerative colitis, or antibiotic-associated colitis.
Drug Interactions
Dapsone topical
May cause temporary orange-yellow skin discoloration when used with benzoyl peroxide.
Erythromycin topical
Concurrent use should be avoided as it may antagonize the effects of clindamycin.
Sulfonamide-containing products
May lead to temporary skin discoloration with benzoyl peroxide.
Other irritating topical agents (e.g., retinoids, salicylic acid)
Increased risk of skin irritation; use with caution or avoid concurrent use.
Storage
Store below 30°C in a cool, dry place, protected from light. Do not freeze. Keep out of reach of children.
Overdose
Topical overdose is unlikely to produce systemic effects. Excessive application may result in redness, peeling, or discomfort. In case of accidental ingestion, seek medical advice immediately.
Pregnancy & Lactation
Use during pregnancy only if the potential benefit justifies the potential risk to the fetus. It is unknown if clindamycin or benzoyl peroxide are excreted in human milk after topical administration. Use with caution in breastfeeding mothers. Consult a doctor.
