Uses & Indications
Dosage
Adults
For Vitamin D deficiency prevention, typically 600-800 IU daily. For treatment, higher doses may be prescribed by a physician based on serum 25(OH)D levels. Usually, 1 capsule once daily.
Elderly
Similar to adults, sometimes higher doses (e.g., 800-1000 IU daily) due to reduced skin synthesis and impaired absorption.
Renal_impairment
Use with caution and monitor serum calcium and phosphate levels. Doses may need adjustment based on severity and concomitant therapies.
How to Take
Take orally, preferably with the largest meal of the day to enhance absorption, as it is a fat-soluble vitamin. Do not crush or chew the capsule; swallow it whole with water.
Mechanism of Action
Cholecalciferol is converted in the liver to 25-hydroxycholecalciferol (calcifediol) and then in the kidneys to 1,25-dihydroxycholecalciferol (calcitriol), the active form of Vitamin D. Absorption

Metabolism
Initially hydroxylated in the liver by CYP2R1 and CYP27A1 to 25-hydroxycholecalciferol [25(OH)D]. This is then further hydroxylated in the kidneys by CYP27B1 to the active form, 1,25-dihydroxycholecalciferol [1,25(OH)2D] (calcitriol).
