Description
Overview of the medicine
Daflon 450 mg is a venotonic and vasoprotective agent used to treat venous insufficiency (e.g., heavy legs, pain, nocturnal cramps, edema) and hemorrhoids (acute and chronic). It is a micronized purified flavonoid fraction composed of 405 mg Diosmin and 45 mg Hesperidin.
Uses & Indications
Dosage
Adults
Venolymphatic insufficiency: 1 tablet twice daily, morning and evening, with meals. Acute hemorrhoidal attacks: 3 tablets twice daily for the first 4 days, then 2 tablets twice daily for the next 3 days, with meals. Chronic hemorrhoidal disease: 1 tablet twice daily with meals.
Elderly
No specific dose adjustment is generally required for elderly patients, but use with caution and monitor for side effects.
Renal_impairment
No specific dosage adjustment guidelines are available; however, use with caution in patients with severe renal impairment due to limited excretion data.
How to Take
Oral administration. Tablets should be taken with meals.
Mechanism of Action
Daflon enhances venous tone by prolonging noradrenaline activity on venous walls, reducing venous distensibility and stasis. It also reduces capillary hyperpermeability and increases lymphatic drainage, leading to a reduction in edema and inflammation.
Pharmacokinetics
Onset
Clinical effects may be observed within a few days to weeks depending on the indication.
Excretion
Mainly through feces (about 80%), and a small percentage (about 14%) via urine.
Half life
Approximately 11 hours (for diosmin, after metabolism).
Absorption
Rapidly absorbed after oral administration. Extensive metabolism leads to phenolic acids which are eliminated.
Metabolism
Extensively metabolized in the liver to phenolic acids, predominantly m-hydroxyphenylpropionic acid.
Side Effects
Contraindications
- Hypersensitivity to the active substance or to any of the excipients.
Drug Interactions
Anticoagulants (e.g., Warfarin)
Theoretical risk of increased bleeding due to potential effect on platelet aggregation. Monitor INR if co-administered.
Storage
Store below 30°C in a dry place, away from direct light and moisture. Keep out of reach of children.
Overdose
Limited data on overdose. Symptoms typically include gastrointestinal disturbances (diarrhea, nausea, abdominal pain) and skin reactions (rash, pruritus). Treatment is symptomatic.
Pregnancy & Lactation
Pregnancy: Studies in animals have not shown teratogenic effects. Limited human data suggest no adverse effects. Use during pregnancy only if clearly needed and under medical supervision. Lactation: It is not known whether the active substance is excreted in human milk. A decision must be made whether to discontinue breastfeeding or to discontinue/abstain from Daflon therapy.
