Description
Overview of the medicine
Debion-100 mg Tablet contains Thiamine Hydrochloride, a water-soluble vitamin (B1), essential for carbohydrate metabolism and nerve function. It is used to prevent and treat Thiamine deficiency.
Uses & Indications
Dosage
Adults
For deficiency, 10-100 mg daily. For severe deficiency (Wernicke-Korsakoff syndrome), higher doses may be given parenterally under medical supervision.
Elderly
No specific dose adjustment generally required, but caution with polypharmacy and renal/hepatic impairment.
Renal_impairment
No specific dose adjustment recommended, as it is primarily excreted renally and considered safe.
How to Take
Oral. Can be taken with or without food. Swallow the tablet whole with water.
Mechanism of Action
Thiamine is converted to thiamine pyrophosphate (TPP), a coenzyme for several metabolic enzymes, including pyruvate dehydrogenase and alpha-ketoglutarate dehydrogenase, crucial for carbohydrate metabolism and energy production.
Pharmacokinetics
Onset
Rapid, effects seen within hours to days depending on severity of deficiency.
Excretion
Excreted primarily in the urine as inactive metabolites or unchanged thiamine, especially after high doses.
Half life
Approximately 9-18 days in humans (for TPP).
Absorption
Well absorbed from the gastrointestinal tract, primarily in the duodenum, through an active transport mechanism at lower concentrations and passive diffusion at higher concentrations.
Metabolism
Metabolized in the liver to active thiamine pyrophosphate (TPP) and other inactive metabolites.
Side Effects
Contraindications
- Hypersensitivity to thiamine or any component of the formulation.
Drug Interactions
Fluorouracil
May inhibit thiamine phosphorylation, potentially leading to thiamine deficiency when co-administered.
Oral Contraceptives
Some studies suggest long-term use might slightly increase thiamine requirements, but clinical significance is usually minimal.
Thiaminase-containing foods
Consumption of large amounts of raw fish, shellfish, tea, and coffee may interfere with thiamine absorption or increase its degradation.
Storage
Store below 30°C in a dry place, protected from light and moisture. Keep out of reach of children.
Overdose
Thiamine is water-soluble, and excessive amounts are generally excreted in urine. Overdose is rare with oral administration and usually causes no significant adverse effects. Very high parenteral doses might rarely cause flushing, weakness, peripheral vasodilation, and anaphylaxis.
Pregnancy & Lactation
Generally considered safe and often recommended as part of prenatal vitamins. Thiamine is excreted in breast milk in small amounts; safe for nursing mothers at recommended doses.

WHO Essential Medicine
YesClinical Trials
Extensive historical data supports the efficacy of Thiamine. Modern clinical trials often focus on specific patient populations with deficiencies (e.g., alcoholics, bariatric surgery patients) or exploring new formulations and delivery methods.
Lab Monitoring
- Thiamine levels in blood (for diagnosis of deficiency)
- Erythrocyte transketolase activity (functional test for thiamine status)
Doctor Notes
- Crucial for patients with chronic alcoholism, malabsorption syndromes (e.g., Crohn's disease, celiac disease), or post-bariatric surgery.
- In suspected Wernicke's encephalopathy, administer thiamine before glucose infusion to prevent worsening of neurological symptoms.
- Patients undergoing hemodialysis may require increased thiamine supplementation.
Patient Guidelines
- Take as directed by your healthcare provider, typically once daily.
- Do not exceed the recommended dose without consulting a doctor.
- Store at room temperature, away from moisture and direct heat, and out of reach of children.
Missed Dose Advice
Take the missed dose as soon as you remember. If it is almost time for your next dose, skip the missed dose and resume your regular dosing schedule. Do not double dose to make up for a missed one.
Driving Precautions
No specific precautions are generally needed as Debion-100 mg Tablet does not typically impair driving ability or operating machinery.
Lifestyle Advice
- Maintain a balanced diet rich in thiamine (found in whole grains, meat, legumes, nuts, and yeast).
- Limit alcohol intake, especially if you have a thiamine deficiency or are at risk.
