Description
Overview of the medicine
Demax 0.5 mg tablet contains Clonazepam, a benzodiazepine, used to treat anxiety disorders, panic disorder, and certain types of seizures. It works by enhancing the activity of a neurotransmitter called gamma-aminobutyric acid (GABA) in the brain.
Uses & Indications
Dosage
Adults
Initial dose for panic disorder: 0.25 mg twice daily; may increase to 1 mg/day after 3 days. For seizure disorders: Initial dose 0.5 mg three times daily, gradually increased by 0.5-1 mg every 3 days until seizure control is achieved (maximum 20 mg/day).
Elderly
Lower doses are recommended due to increased sensitivity and risk of adverse effects. Initial dose of 0.125 mg once or twice daily.
Renal_impairment
No specific dosage adjustment guidelines for renal impairment; use with caution.
How to Take
Take orally with or without food. Swallow the tablet whole with water. Do not crush or chew.
Mechanism of Action
Clonazepam enhances the effect of gamma-aminobutyric acid (GABA) at GABA-A receptors, leading to increased chloride ion influx into neurons, hyperpolarization, and reduced neuronal excitability in the central nervous system.
Pharmacokinetics
Onset
30-60 minutes
Excretion
Mainly excreted in the urine as metabolites, with a small amount excreted in feces.
Half life
18-50 hours (average 30-40 hours)
Absorption
Rapidly and completely absorbed after oral administration. Peak plasma concentrations are reached within 1-4 hours.
Metabolism
Primarily metabolized in the liver by nitroreduction to 7-aminoclonazepam and by N-acetylation.
