Diatrol 80 mg Tablet contains Gliclazide, an oral hypoglycemic agent from the sulfonylurea class, used to control high blood sugar levels in adults with type 2 diabetes mellitus. It stimulates insulin release from the pancreatic beta cells.
Uses & Indications
Dosage
Adults
Initial dose: 40-80 mg once daily, preferably with breakfast. Gradually increase if needed, up to a maximum of 320 mg/day in divided doses. Standard maintenance dose: 80-160 mg/day.
Elderly
Initial dose 40 mg once daily, adjusted carefully based on glycemic response and renal function.
Renal_impairment
Mild to moderate renal impairment: Usually no dosage adjustment needed, but careful monitoring is advised. Severe renal impairment: Not recommended, or use with extreme caution at reduced doses.
How to Take
Oral administration. Take preferably with breakfast or the first main meal of the day. Swallow the tablet whole with water; do not crush or chew. Adhere to diet and exercise recommendations.
Mechanism of Action
Gliclazide works by stimulating the beta cells in the pancreas to release more insulin. It binds to the sulfonylurea receptor (SUR1) on the pancreatic beta-cell membrane, leading to the closure of ATP-sensitive potassium channels, membrane depolarization, calcium influx, and subsequent insulin secretion.

Contraindications
- Hypersensitivity to gliclazide or other sulfonylureas or any excipients.

Drug Interactions
Miconazole
Potentiates hypoglycemic effect, leading to severe hypoglycemia. Concomitant use is contraindicated.
Phenylbutazone, Alcohol, Sulfonamides, NSAIDs, Beta-blockers
May potentiate the hypoglycemic effect of gliclazide.
Corticosteroids, Diuretics, Barbiturates, Estrogens, Progestogens
May reduce the hypoglycemic effect of gliclazide.
Storage
Store in a cool, dry place below 30°C. Protect from light and moisture. Keep out of reach of children.
Overdose
Symptoms include severe hypoglycemia (e.g., sweating, trembling, confusion, unconsciousness). Management involves immediate glucose administration (oral glucose for conscious patients, intravenous glucose or glucagon for unconscious patients) and monitoring for at least 24-48 hours due to prolonged hypoglycemic effects.
Pregnancy & Lactation
Contraindicated during pregnancy and breastfeeding. Insulin is the preferred treatment for diabetes in pregnant women. Consult a doctor for alternative management.
