Overview of the medicine
Doxibac 100 mg Capsule contains Doxycycline, a broad-spectrum tetracycline antibiotic used to treat various bacterial infections including respiratory tract infections, urinary tract infections, skin infections, sexually transmitted infections, and certain parasitic infections. It works by stopping the growth of bacteria.
Uses & Indications
Dosage
Adults
Typical dose is 100 mg every 12-24 hours depending on the infection. For severe infections, 200 mg on the first day, then 100 mg daily.
Elderly
No specific dose adjustment is usually required, but caution is advised in patients with hepatic impairment.
Renal_impairment
No dosage adjustment is necessary for patients with impaired renal function, as doxycycline is primarily eliminated via feces.
How to Take
Take with a full glass of water, preferably in an upright position (sitting or standing) to prevent esophageal irritation/ulceration. Can be taken with food or milk to reduce gastrointestinal upset, though absorption may be slightly reduced. Do not lie down for at least 30 minutes to 1 hour after taking the capsule.
Mechanism of Action
Doxycycline exerts its antimicrobial effect by inhibiting bacterial protein synthesis. It reversibly binds to the 30S ribosomal subunit, blocking the binding of aminoacyl tRNA to the mRNA-ribosome complex, thereby preventing the addition of amino acids to the growing peptide chain.

Onset
Antimicrobial effects usually begin within 1-2 hours of administration.
Excretion
Mainly excreted unchanged in the feces via intestinal secretion (non-biliary) and to a lesser extent in the urine.
Half life
Approximately 16-22 hours, allowing for once or twice daily dosing.
Absorption
Well-absorbed orally, approximately 90-100% bioavailability. Food and dairy products may slightly reduce absorption, but usually not clinically significant.
Metabolism
Partially metabolized in the liver, but the primary route of elimination is not metabolic.
Side Effects
Contraindications
- Hypersensitivity to doxycycline or any other tetracycline antibiotic.
- Pregnancy (especially during the second and third trimesters) due to potential tooth discoloration and skeletal effects in the fetus.
- Lactation, as doxycycline is excreted in breast milk.
- Children under 8 years of age (due to permanent tooth discoloration and enamel hypoplasia).
Drug Interactions
Warfarin
Doxycycline may potentiate the anticoagulant effect of warfarin, requiring INR monitoring and potential dose adjustment.
Penicillins
Tetracyclines can interfere with the bactericidal action of penicillins. Avoid concomitant use.
Oral Contraceptives
Tetracyclines may reduce the effectiveness of oral contraceptives, advise using an alternative or additional method of birth control.
Antacids, iron preparations, calcium supplements
Can decrease doxycycline absorption. Take doxycycline 1-2 hours before or 2-3 hours after these products.
Storage
Store in a cool, dry place, below 30°C. Protect from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose may include nausea, vomiting, and diarrhea. In case of overdose, discontinue the drug and institute symptomatic and supportive therapy. Gastric lavage may be considered. Hemodialysis is not effective in removing doxycycline from the system.
Pregnancy & Lactation
Doxycycline is categorized as Pregnancy Category D. It can cause permanent discoloration of teeth and inhibition of bone growth in the fetus. It is contraindicated during pregnancy. It is excreted in breast milk and should be avoided during breastfeeding due to potential effects on the infant's teeth and bones.
