Description
Overview of the medicine
Desloratadine is a non-drowsy antihistamine used to relieve allergy symptoms such as sneezing, runny nose, itchy eyes, and hives. It is effective in treating symptoms of seasonal and perennial allergic rhinitis, and chronic idiopathic urticaria.
Uses & Indications
Dosage
Adults
5 mg once daily.
Elderly
No dosage adjustment generally required, but caution in patients with renal or hepatic impairment. A starting dose of 5 mg every other day may be considered if needed.
Renal_impairment
For patients with severe renal impairment (creatinine clearance <30 mL/min), a starting dose of 5 mg every other day is recommended.
How to Take
Administer orally, with or without food. Swallow the tablet whole with water.
Mechanism of Action
Desloratadine is a long-acting, non-sedating, selective peripheral H1-receptor antagonist. It prevents the action of histamine, a substance produced by the body that causes allergic symptoms, by blocking H1 receptors.

Primarily excreted in urine and feces as conjugated metabolites.

Approximately 27 hours.
Absorption
Rapidly absorbed after oral administration. Peak plasma concentrations are reached in approximately 3 hours.

Extensively metabolized in the liver to 3-hydroxydesloratadine, an active metabolite, via hydroxylation and subsequent conjugation.

Contraindications
- Hypersensitivity to desloratadine, loratadine, or any component of the formulation.
Drug Interactions
Alcohol
No potentiation of alcohol-induced impairment has been observed.
Cimetidine
May increase plasma concentrations of desloratadine.
Fluoxetine
May increase plasma concentrations of desloratadine.
Azithromycin
May increase plasma concentrations of desloratadine.
Erythromycin
May increase plasma concentrations of desloratadine.
Ketoconazole
May increase plasma concentrations of desloratadine.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose may include somnolence, tachycardia, and headache. In case of overdose, general symptomatic and supportive measures should be instituted. Desloratadine is not eliminated by hemodialysis.
Pregnancy & Lactation
Pregnancy Category C. Use during pregnancy only if the potential benefit justifies the potential risk to the fetus. Desloratadine is excreted into breast milk; a decision should be made whether to discontinue nursing or to discontinue the drug, taking into account the importance of the drug to the mother.
